中文摘要: 该试验旨在建立一种高效液相色谱法(high performance liquid chromatography,HPLC)检测蛋鸡饲喂白藜芦醇后获得的试验鸡蛋中白藜芦醇富集情况的方法。使用C18(4.6 mm×250 mm,5 μm)色谱柱,以乙酸+水(0.9 mL+750 mL)∶乙腈=65∶35(体积比)为流动相;流速 1.0 mL/min;检测波长 306 nm;柱温 30 ℃;进样量 10 μL。结果表明:白藜芦醇在线性范围1 μg/mL~10 μg/mL内标准品浓度与峰面积线性关系良好,标准曲线方程为y=72.777x,相关系数R2=0.999 7,平均加标回收率为86.72%~99.41%,RSD范围为0.5%~1.05%;使用该方法对鸡蛋蛋黄中白藜芦醇进行检测,结果显示,普通鸡蛋组蛋黄中不含白藜芦醇,添加1.5 g/kg白藜芦醇组和3 g/kg白藜芦醇组蛋黄中白藜芦醇含量分别为0.083 μg/g和0.371 μg/g。该方法精密度较高,出峰良好,与杂峰完全分离,适用于试验鸡蛋中的白藜芦醇的测定。
Abstract:The objective of this study was to establish a high performance liquid chromatography(HPLC)method to quantitate resveratrol enrichment in experimental eggs produced by laying hens whose diet was supplemented with resveratrol.A C18(4.6 mm×250 mm,5 μm)column was used with acetic acid+water(0.9 mL+750mL)∶acetonitrile=65∶35(volume ratio)as mobile phase.Flow rate was 1.0 mL/min.Detection wavelength was 306 nm,and column temperature was 30℃.Injection volume was 10 μL.A good linear relationship was observed between peak area and resveratrol standard concentrations,with a linear range of 1 μg/mL-10 μg/mL.The standard curve equation was y=72.777x,and the correlation coefficient was R2=0.999 7.Average recoveries were 86.72%-99.41%,and RSD range was 0.5%-1.05%.No resveratrol was detected in the yolks of the normal egg group.Resveratrol content in the yolks of the 1.5 g/kg and 3 g/kg resveratrol groups were 0.083 μg/g and 0.371 μg/g,respectively.The method was accurate,and suitable for quantitation of resveratrol in experimental eggs.
