中文摘要: 该研究旨在探讨桑叶生物碱(mulberry leaf alkaloids,MLA)对高果糖代谢综合征小鼠糖脂代谢紊乱的影响。建立高果糖饮食诱导的代谢综合征小鼠模型,以MLA 40、80、160 mg/(kg·d)三个剂量进行干预,检测糖脂代谢相关生化指标,观察小鼠肝脏组织结构,研究MLA对小鼠氧化应激和炎症的影响。结果显示,模型组小鼠与正常组小鼠相比,体重显著增高,空腹血糖虽无显著性差异,但葡萄糖的耐受能力明显降低,肝脏组织结构受损,血清甘油三酯(triglyceride,TG)、总胆固醇(total cholesterol,TC)、谷丙转氨酶(alanine aminotransferase,ALT)、谷草转氨酶(aspartate transaminase,AST)、丙二醛(malondialdehyde,MDA)和肿瘤坏死因子 α(tumor necrosis factor α,TNF-α)、白细胞介素-1β(interleukin-1β,IL-1β)和白细胞介素-6(interleukin-6,IL-6)水平显著升高,血清超氧化物歧化酶(superoxide dismutase,SOD)活力明显下降。MLA的干预可以抑制高果糖导致的体重异常增加,改善其葡萄糖耐受能力,降低血清TC、TG、ALT、AST、MDA水平,提高血清SOD的活力,改善肝脏组织结构病变,降低炎症因子TNF-α、IL-1β和IL-6的水平。这些结果证实MLA能够改善高果糖代谢综合征小鼠糖脂代谢紊乱,保护肝脏,其作用可能与MLA能够改善机体抗氧化能力和减轻炎症反应有关。
Abstract:The study was conducted to investigate the effect of mulberry leaf alkaloids(MLA)on glucose and lipid metabolism disorders in mice with a metabolic syndrome induced by a high fructose diet.An animal model of metabolic syndrome in high-fructose mice was established.Three doses of MLA[40 mg/(kg·d),80 mg/(kg·d)and 160 mg/(kg·d)]were administered.The biochemical indexes related to glucose and lipid metabolism were monitored,and the liver tissue structure was studied.Additionally,the effects of MLA on oxidative stress and inflammation in mice were investigated.The results showed that compared with the control group,the body weight of the model group increased significantly.The fasting blood glucose levels showed no significant difference,but glucose tolerance was significantly reduced,and the liver tissue structure was damaged.Serum triglyceride(TG),total cholesterol(TC),alanine aminotransferase(ALT),aspartate transaminase(AST),malondialdehyde(MDA),tumor necrosis factor α(TNF-α),interleukin-1β(IL-1β),and interleukin-6(IL-6)were significantly increased.The activity of superoxide dismutase(SOD)was significantly decreased.MLA intervention could in-hibit abnormal weight gain caused by a high fructose diet;improve glucose tolerance;reduce serum TC,TG,ALT,AST,MDA levels;increase serum SOD activity;improve the pathological damage to liver tissue;and reduce the levels of inflammatory factors TNF-α,IL-1β,and IL-6.These results confirmed that MLA could improve the glucose and lipid metabolism disorder induced by a high fructose diet in mice.The protective effect of MLA may be attributed to the improvement in antioxidant capacity and inhibition of inflammatory response.
