中文摘要: 以乙醇为提取剂,通过冷凝回流、盐酸酸化、乙酸乙酯萃取提取枸杞中酚酸成分,优化并建立宁夏枸杞中9种酚酸化合物的高效液相色谱检测方法。采用Ultimatepagenumber_ebook=156,pagenumber_book=148 LP-C1(8250 mm×4.6 mm,5 μm)色谱柱,以乙腈-0.1%磷酸水溶液为流动相进行梯度洗脱,在流速1.0 mL/min、检测波长为280 nm、柱温30℃条件下对没食子酸、香豆酸、原儿茶醛、儿茶素、绿原酸、咖啡酸、表儿茶素、丁香酸、阿魏酸进行分离,9种酚酸在46 min内完全分离。各组分的质量浓度与其峰面积具有良好的线性关系,相关系数均大于0.993 0,方法的平均回收率在73.6%~99.8%,相对标准偏差小于8%。该方法可用于枸杞中酚酸化合物含量的测定,能够为枸杞质量控制提供一定技术支撑。
Abstract:Phenolic acid compounds are typically extracted by condensation reflux,hydrochloric acid acidification,and ethyl acetate extraction.The present study sought to establish and optimize a reversed phase high-performance liquid chromatography method for the simultaneous determination of phenolic constituents of Lycium barbarum L.Chromatography was performed using an Ultimatepagenumber_ebook=156,pagenumber_book=148 LP-C18column(250 mm×4.6 mm,5 μm)with gradient elution using a mobile phase composed of acetonitrile-0.1% phosphoric acid,flow rate of 1.0 mL/min,and column temperature of 30°C.The absorbance of the eluted fractions was measured at 280 nm.Nine kinds of phenolic acids—gallic acid,coumaric acid,original tea aldehyde,catechin,chlorogenic acid,coffee acid,benzoic acid,epicatechin,and ferulic acid—were completely separated within 46 min.A strong positive linear correlation was evident between the concentration and peak area.The correlation coefficient was greater than 0.993 0.The average recoveries ranged from 73.6% to 99.8%,with relative standard deviations being less than 8%.The described method could be applied to the quantitative and qualitative analyses of phenolic acids in Lycium barbarum L..
