中文摘要: 该研究是开发一种准确、快速的方法来测定肉和肉制品中丙二醛的含量。以三氯乙酸(trichloroacetic acid,TCA)萃取、2-硫代巴比妥酸(2-thiobarbituric acid,TBA)反应作为基础,用超高效液相色谱-荧光检测器(ultraperformance liquid chromatography-fluorescence detector,UPLC-FLD)及超高效液相色谱-二极管阵列检测器(ultraperformance liquid chromatography-diode array detector,UPLC-DAD)进行肉及肉制品中丙二醛(malondialdehyde,MDA)的定量分析。通过实际样品的分析测定结果可知,除了煮熟火腿和法兰克福香肠外,两种UPLC方法测定的大多数肉制品中的MDA含量相近。而硫代巴比妥酸反应物(thiobarbituric acid reactive substances,TBARS)分光光度法检测的MDA含量比实际含量偏高。因此,采用这两种色谱方法,特别是UPLC-FLD法比传统的TBARS方法更准确,可避免检测的肉制品中MDA含量偏高。
Abstract:The purpose of the study was to develop an accurate and rapid method for the determination of malondialdehyde(MDA)in meat and meat products.Based on trichloroacetic acid(TCA)extraction and 2-thiobarbituric acid (TBA)reaction,quantitative analysis of MDA in meat and meat products was conducted using ultra-performance liquid chromatography with fluorescence detection(UPLC-FLD)and ultra-performance liquid chromatography with diode array detection(UPLC-DAD)methods.The results showed that the content of MDA in most meat products was similar,except in cooked ham and Frankfurt sausage,when determined by the two UPLC methods.However,the content of MDA detected through thiobarbituric acid reactive substances(TBARS)spectrophotometry was higher than the actual thiobarbituric acid reactant content.Therefore,these two chromatographic methods,especially the UPLC-FLD method,were more accurate than the traditional TBARS method.They enabled accurate detection of the MDA content in meat products and helped avoid the use of the contaminated meat.
