中文摘要: 利用多重基因表达(GenomeLabTMeXpress Profiling,GeXP)遗传分析系统建立一种可单管一次同时检测大肠杆菌O157∶H7、金黄色葡萄球菌、志贺氏菌、副溶血性弧菌、单核细胞增生李斯特菌和沙门氏菌6种肉及肉制品中致病菌的多重聚合酶链式反应(polymerase chain reaction,PCR)检测方法。评价此方法特异性和灵敏性,并初步应用于市场随机购买的肉类样品的检测。结果显示,该方法可在短时间内实现同时对上述6种致病菌进行检测,且检测灵敏度低至103CFU/mL,为食源性致病菌高通量快速有效检测提供新的方法技术支持,具有较强的实际应用价值。

Abstract:A single-tube GenomeLabTM eXpress Profiling (GeXP)-based multiplex polymerase chain reaction(PCR)assay for simultaneous detection of six common pathogens (Escherichia coli O157∶H7,Staphylococcus aureus,Shigella,Vibrio parahaemolyticus,Listeria monocytogenes,and Salmonella) in meat and meat products was established.The specificity and sensitivity of the GeXP multiplex PCR assay were evaluated,and further validated the optimized assay by pathogen detection in random meat samples obtained from a local market.The GeXP multiplex PCR established in this study successfully detected six common foodborne pathogens simultaneously,with good specificity and high sensitivity(as low as 103CFU/mL).This method could improve rapid and effective high-throughput detection of foodborne pathogens,provide needed technical support for foodborne disease detection,and achieve favorable prospective application.
