中文摘要: 为鉴别食品中虹鳟源性成分,以线粒体ATP6 基因为靶点,设计虹鳟特异性聚合酶链式反应(polymerase chain reaction,PCR)的引物、探针,利用实时荧光聚合酶链式反应(real-time fluorescent PCR,RT-PCR)技术,建立针对虹鳟源性成分的实时荧光PCR 快速检测方法;选取17 种市场上常见鱼类进行试验,结果均无扩增反应,同时通过DNA灵敏度试验,表明该研究建立的鉴别方法DNA 浓度灵敏度能达到0.1 ng/μL,且特异性良好。同时,利用该方法对市售的22 种商品三文鱼及其制品进行检测,结果显示10 种商品中含有虹鳟(Oncorhynchus mykiss)成分。该研究所建立的检测方法可用于对市售产品中虹鳟源性成分的鉴别,同时也表明虹鳟是市售三文鱼的主要鱼种之一。
Abstract:To identify rainbow trout(Oncorhynchus mykiss)-derived components in food,the ATP6 mitochondrial gene was selected as the target to design rainbow trout-specific primers and probes for polymerase chain reaction (PCR).Real-time fluorescent PCR enabled the rapid identification of rainbow trout-derived components.The assay was used for 17 common commercially available fish species.No amplification reaction was observed. The DNA concentration sensitivity was 0.1 ng/μL and was accompanied by good specificity. The assay was also used to test 22 commercial salmon and their products. Ten of these commodities contain rainbow trout ingredients.The developed detection method can be applied to identify rainbow trout components in commercial products.Rainbow trout was one of the main species of some commercially available salmon.
