南极磷虾油改善衰老小鼠抗氧化能力的作用研究

中文摘要: 研究南极磷虾油对衰老小鼠抗氧化能力的改善作用。以雄性ICR 小鼠为研究对象,采用皮下注射D-半乳糖(500 mg/kg bw)方法建立小鼠衰老模型,正常组注射等体积生理盐水,造模6 周后,衰老模型小鼠根据血清丙二醛(malondialdehyde,MDA)水平分为模型组、不同剂量南极磷虾油组(50、150、300 mg/kg bw),各南极磷虾油剂量组每日灌胃给予相应剂量南极磷虾油,正常组和模型组灌胃等体积大豆油,连续灌胃30 d 后,测定小鼠血清MDA 含量、超氧化物歧化酶(superoxide dismutase,SOD)活力、蛋白质羰基含量及全血还原型谷胱甘肽(reduced glutathione,GSH)含量。结果表明,模型组小鼠较正常组血清MDA 和蛋白质羰基含量显著升高(P<0.05)、血清SOD 活力和全血GSH 水平显著降低(P<0.01)。与模型组小鼠相比,南极磷虾油各剂量组均能提高血清SOD 活力和全血GSH 水平、降低血清MDA 和蛋白质羰基含量,且中、高剂量组效果显著(P<0.05)。南极磷虾油可保护D-半乳糖所致衰老小鼠的氧化损伤,提高其抗氧化能力,起到延缓衰老的作用。
Abstract:The stimulatory effect of Antarctic krill oil on cellular antioxidant capacity in D-galactose-induced aging mice was evaluated in the present study. Male ICR mice were injected subcutaneously with D-galactose(500 mg/kg bw)to establish the aging model. The normal group was injected with an equal volume of normal saline.Six weeks post injection,the aging model mice were divided into a vehicle treated group,and three groups each receiving a different dose of Antarctic krill oil (50,150 and 300 mg/kg bw).Mice were sorted into groups in a way that equilibrated initial serum malondialdehyde(MDA)distributions between groups.The groups receiving different doses of Antarctic krill oil received their daily doses of krill oil by oral gavage. The healthy group and the vehicle-treated aging model group were given equal volumes of soybean oil by oral gavage.After 30 days of continuous administration,MDA levels,superoxide dismutase(SOD)activities,and protein carbonyl levels in serum,and reduced glutathione(GSH)levels in whole blood were determined. Compared with the healthy group,serum MDA and protein carbonyl levels in the vehicle-treated aging model group were significantly increased(P<0.05),while serum SOD activities and whole blood GSH levels were significantly decreased(P<0.01).Relative to the model group,intervention with Antarctic krill oil increased serum SOD activities and whole blood GSH levels,and reduced serum MDA and protein carbonyl levels in aging model mice.The protective effects seen in the medium and high dose groups were significant (P<0.05). Antarctic krill oil could efficiently ameliorate oxidative damage caused by D-galactose,and improve cellular antioxidant capacities in aging mice.
