延衰组合物对酵母细胞寿命及氧化应激的研究

中文摘要: 以西洋参、五味子、麦冬、茯苓、葛根及这5 种原料按特定比例组成的组合提取物为研究对象,利用酵母细胞的时序寿命(chronological lifespan,CLS)优选延衰活性物质,进而检测优选出的干预组酵母细胞内活性氧(reactive oxygen species,ROS)水平、线粒体膜电位(mitochondrial membrane potential,MMP)及与氧化应激和碳水化合物积累密切相关的转录因子Msn2/4、Gis1 和Hsf1 的激活情况。结果表明:与各单一原料提取物相比,组合提取物可显著延长酵母细胞的时序寿命(p<0.05)。与空白对照组的相对荧光强度值(100%)相比,组合提取物组酵母细胞内的活性氧水平(82.17±2.48)%(p<0.05)和线粒体膜电位(54.44±1.23)%均显著降低(p<0.001)。此外,组合提取物还可显著激活酵母细胞内与氧化应激和碳水化合物积累密切相关的Msn2/4、Gis1 和Hsf1 转录因子[其红色相对荧光强度值为(151.85±3.50)%;绿色相对荧光强度值为(154.44±5.13)%,p<0.001]。表明组合提取物对酵母具有延长寿命的作用,且能改善衰老过程中氧化应激和能量代谢问题。
Abstract:The anti-aging activities of Panax quinquefolius,Schisandra chinensis,Ophiopogon japonicus(Linn.f.) Ker-Gawl., Wolfiporia extensa(Peck)Ginns, and lobed kudzuvine root extract,and extracts of these five herbs combined in specific proportions were investigated using Saccharomyces cerevisiae as a model. The extract combination was able to significantly prolong the lifespan(CLS)of yeast cells,while the individual extracts had no such effect.The extract combination also significantly reduced reactive oxygen species(ROS)to(82.17±2.48)%(p<0.05)and decreased mitochondrial membrane potential(MMP)to (54.44±1.23)%(p<0.001)relative to vehicle treated cells(relative fluorescence values:100%).Moreover,the extract combination significantly increased the expression/activity of transcription factors Msn2/4,Gis1,and Hsf1[relative red fluorescence value:(151.85 ± 3.50)%; relative green fluorescence value:(154.44 ± 5.13)%,p < 0.001] which correlate strongly with the oxidative-stress response and carbohydrate accumulation. In summary,the anti-aging extract combination could prolong the lifespan of yeast cells,ameliorate aging-associated oxidative stress,and enhance energy metabolism.
