中文摘要: 为制备高活性屎肠球菌R2纯种酸浆发酵剂,首先对屎肠球菌R2的有氧呼吸代谢进行验证,随后以600 nm下的光密度值为测定指标对屎肠球菌R2有氧呼吸增殖的培养温度、血红素浓度、初始pH值、摇床转速进行四因素三水平单因素和正交试验优化,获得该菌株有氧呼吸最佳增殖条件;在此基础上采用冷冻干燥制备酸浆发酵剂,并对发酵剂的品质进行评价。结果表明,屎肠球菌R2有氧呼吸最佳增殖条件为,温度32℃,转速175 r/min,血红素浓度3.0 μg/mL,初始pH 5.9;采用有氧呼吸增殖得到的酸浆发酵剂活菌数为1.33×109CFU/g,且其产酸性能良好,可用于酸浆的加工。
Abstract:The purpose of this study was to optimize the conditions for preparation of high-activity Enterococcus faecium R2 acid pulp starters.After verifying aerobic respiration,optimization of aerobic respiration by single factor and orthogonal tests of temperature,heme concentration,initial pH,and rotation speed was performed by measuring optical density at 600 nm.The quality of acid pulp starters prepared by freeze-drying was then evaluated.The optimal proliferation conditions for E.faecium R2 undergoing aerobic respiration were: temperature,32 ℃;rotation speed,175 r/min;heme concentration,3.0 μg/mL,and initial pH 5.9.The number of viable bacteria in acid pulp starters cultivated with aerobic respiration reached 1.33×109CFU/g.Its acid production was good,and could be used in the processing of acid pulp.
