中文摘要: 以水翁花为原材料,利用单因素及响应面优化纤维素酶法提取总黄酮工艺条件,并考察总黄酮提取物的抗氧化活性。结果表明,当酶用量0.56%、酶解温度56℃、酶解时间61 min、pH 4.9时,总黄酮提取率为(3.85±0.09)%,与预测值(3.87±0.08)%吻合得很好。当总黄酮溶液浓度为0.75 mg/mL时,其对ABTS+自由基的清除率达到(70.6±3.0)%;当浓度为1.6 mg/mL时,对油脂氧化的清除率为(58.7±2.4)%;当总黄酮溶液浓度为0.75 mg/mL时,测得700 nm处的吸光度值为(0.60±0.03)。研究表明,水翁花总黄酮具有较好的抗氧化活性。
Abstract:With the buds of Cleistocalyx operculatus as raw material,single factor and response surface methodology were used to optimize the cellulase assisted extraction of total flavonoids.The antioxidant activity of total flavonoids extract was also evaluated.The results showed that the optimal conditions were cellulase dosage 0.56%,hydrolysis temperature 56℃,hydrolysis time 61 min,and pH 4.9.Under the optimum conditions,the extraction yield of total flavonoids was (3.85±0.09)%,which was in good agreement with the predicted value of(3.87±0.08)%.When the concentration of total flavonoids was 0.75 mg/mL,the free radical scavenging rate of total flavonoids on ABTS+free radical was(70.6±3.0)%.The inhibition rate of oil oxidation was(58.7±2.4)%,when the concentration of total flavonoids was 1.6 mg/mL.When the concentration of total flavonoids was 0.75 mg/mL,the absorbance value at 700 nm was(0.60±0.03).The results obtained in the present study indicated that it has good antioxidant activity.
