中文摘要: 探究银耳多糖、茯苓多糖、香菇多糖、猴头菇多糖和竹荪多糖5种食用菌多糖及其两两复配得到的10种复配多糖对小鼠巨噬细胞RAW264.7的免疫调节作用。将RAW264.7细胞分为正常对照组、阳性组和样品(不同浓度的银耳多糖、茯苓多糖、香菇多糖、猴头菇多糖和竹荪多糖及其10种复配多糖)处理组,通过检测巨噬细胞的细胞活力、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)分泌水平以及吞噬能力评价免疫调节作用。结果表明:与正常对照组相比,在质量浓度5 μg/mL~320 μg/mL范围内,5种食用菌多糖在一定浓度时皆能表现出显著增强巨噬细胞RAW264.7的细胞活力,提高巨噬细胞的吞噬能力,促进细胞因子TNF-α的分泌效果(P<0.05)。此外,10种复配多糖在RAW264.7细胞的细胞活力、TNF-α分泌和吞噬能力皆呈现出显著的促进效果。10种复配多糖中,基于香菇多糖和猴头菇多糖的复配多糖在浓度为160 μg/mL时促进细胞因子TNF-α的分泌效果最好;基于茯苓多糖和香菇多糖复配得到的复配多糖对细胞吞噬功能的增强效果最佳,且优于其组成多糖单独作用效果。基于银耳多糖和竹荪多糖复配得到的复配多糖在浓度为80 μg/mL时促进RAW264.7分泌TNF-α的效果优于其组成多糖单独作用,在浓度为160 μg/mL时对细胞吞噬功能的增强效果优于其组成多糖。银耳、茯苓、香菇、猴头菇、竹荪来源的多糖单独作用及其两两复配得到的复配多糖对RAW264.7细胞均具有免疫调节活性。不同多糖间可能存在相互作用,复配后免疫调节活性优于单独作用时免疫调节活性,例如茯苓多糖和香菇多糖,银耳多糖和竹荪多糖。

Abstract:This study aimed to explore the immunomodulatory effects of polysaccharides derived from five kinds of edible fungi,including Tremella fuciformis,Poria cocos,Lentinus edodes,Hericium erinaceus and Dictyophora indusiata,as well as the compound polysaccharides obtained by mixing these polysaccharides in pairs on the mouse macrophage RAW264.7.RAW264.7 cells were divided into the normal control group,positive control group,and sample treatment(lentinan,polysaccharides derived from Tremella fuciformis,Poria cocos,Hericium erinaceus and Dictyophora indusiata,and ten complexes obtained by mixing the polysac-charides in different concentrations)groups.Macrophage viability and the levels of tumor necrosis factor(TNF)-α were evaluated to examine the immunoregulatory effect as well as the phagocytic ability of RAW264.7 cells following treatment.Compared with the normal control group,polysaccharides derived from the five edible fungi significantly enhanced cell viability,promoted the secretion of the cytokine TNF-α,and improved the phagocytic ability of RAW264.7 cells (P<0.05)with concentrations ranging from 5 μg/mL to 320 μg/mL.In addition,the ten polysaccharide complexes demonstrated significant ability to promote cell viability,TNF-α secretion,and phagocytosis of RAW264.7 cells.The compound polysaccharide composed of lentinan and the polysaccharide derived from Hericium erinaceus were most effective in promoting the secretion of TNF-α at a concentration of 160 μg/mL.The compound polysaccharide composed of lentinan and the polysaccharide derived from Poria cocos demonstrated the strongest enhancement of the phagocytic function of cells;this effect was superior to those of its two components.The compound polysaccharide composed of the polysaccharides derived from Tremella fuciformis and Dictyophora indusiata promoted the secretion of TNF-α by RAW264.7 cells.This effect was stronger than those of its component polysaccharides at a concentration of 80 μg/mL.The ability of the complex to enhance the phagocytic function of cells was stronger than those of its component polysaccharides at a concentration of 160 μg/mL.Polysaccharides derived from Tremella fuciformis,Poria cocos,Lentinus edodes,Hericium erinaceus,and Dictyophora indusiata and their complexes regulated the immune response of the RAW264.7 macrophage line.Different polysaccharides might interact,and the immunoregulatory activities of the compound polysaccharides were superior to those of their component polysaccharides,including lentinan and the polysaccharides derived from Poria cocos,Tremella fuciformis and Dictyophora indusiate.
