中文摘要: 利用超声波辅助技术研究桦褐孔菌多糖的最佳提取工艺,并对其抗氧化活性进行评价。在单因素试验基础上,以多糖提取率为指标,采用Box-Behnken响应面法优化超声辅助提取条件;采用三氯乙酸(trichloroacetic acid,TCA)法纯化多糖后,通过DPPH自由基清除试验来评价其抗氧化活性。结果表明,桦褐孔菌多糖的最佳提取条件为:超声时间31 min,超声温度52℃,液料比为21∶1(mL/g),多糖提取率达到(3.81±0.19)%。桦褐孔菌粗多糖中多糖质量分数为19.0%(以葡萄糖计),蛋白质量分数为13.9%(以牛血清蛋白计)。体外抗氧化试验显示,桦褐孔菌精制多糖对DPPH自由基有一定的清除作用。
Abstract:The optimal ultrasonic-assisted extraction conditions of polysaccharides from Inonotus obliquus was investigated and the antioxidant activity was evaluated.Response surface methodology(RSM)based on Box-Behnken method was used to optimize the ultrasonic-assisted extraction conditions with the yield of polysaccharides as an indicator based on the single-factor tests.The antioxidant activity of polysaccharides was assessed by scavenging assays of DPPH free radicals after purified by trichloroacetic acid(TCA).The results showed that the optimal extraction conditions of polysaccharides were as follows:ultrasonic time 31 min,ultrasonic temperature 52 ℃,liquid/solid ratio 21 ∶1 (mL/g)with the yield of polysaccharides (3.81±0.19)%.The fractions of polysaccharides and proteins in the crude polysaccharides of Inonotus obliquus were 19.0% and 13.9% respectively.The purified polysaccharides from Inonotus obliquus have a certain scavenging effect on DPPH free radical in vitro.
