熟地黄对视网膜损伤小鼠的影响

中文摘要: 采用光损伤小鼠模型,分别用熟地黄水提醇沉提取物和熟地黄水提取物对模型小鼠进行干预,探讨熟地黄对视疲劳的缓解作用。将小鼠分为正常对照组、模型组、熟地黄水提醇沉提取物低剂量组(360 mg/kg)、高剂量组(450 mg/kg)、熟地黄水提取物组(690 mg/kg),共5组,每组6只,对小鼠连续干预8周,再给予光强为(9 000±500)Lux进行照射(7 d),光照结束后用采集晶状体及肝脏进行晶状体抗氧化能力、肝脏抗氧化能力检测。结果表明,通过比较晶状体中谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)和总抗氧化能力(total antioxidant capacity,T-AOC)指标发现,模型组GSH-Px与T-AOC活力均显著低于正常对照组;熟地黄水提醇沉提取物和熟地黄水提取物组GSH-Px与T-AOC活力均明显高于模型组,具有统计学差异(P<0.05),且熟地黄水提醇沉提取物的抗氧化效果随着剂量的升高而升高。从肝脏抗氧化指标结果上看,与模型组比较,熟地黄水提醇沉提取物和熟地黄水提取物均能显著降低丙二醛(malondialdehyde,MDA)含量,提高超氧化物歧化酶(superoxide dismutase,SOD)、过氧化氢酶(catalase,CAT)、GSH-Px活性,差异有统计学意义(P<0.05)。研究结果表明熟地黄提取物对光损伤小鼠晶状体具有较好的保护作用,可对视疲劳起到缓解作用。

Abstract:Taken light-damaged mice as model,the water-extracted alcohol precipitation extract of Radix Rehmanniae Preparata and water extract of Radix Rehmanniae Preparata were used to intervene in model mice,respectively,to explore the mitigating effect of Radix Rehmanniae Preparata on visual fatigue.Mice were divided into normal control group,model group,water-extracted alcohol precipitation extract low-dose group(360 mg/kg),high-dose group (450 mg/kg),and water extract group (690 mg/kg),each group were intervened for 8 weeks,and then irradiated with light intensity of(9 000 ± 500)Lux for irradiation(7 d).After irradiation,the lens and liver were collected to test the antioxidant capacity of the lens and liver.The results showed that by comparing the indexes of glutathione peroxidase(GSH-Px)and total antioxidant capacity(TAOC)in the lens,it was found that the activity of GSH-Px and T-AOC in the model group were significantly lower than that in the normal control group.The GSH-Px and T-AOC activities of the extract group were significantly higher than the model group(P<0.05),and showed a dose-dependent.From the results of liver antioxidant indexes,compared with the model group,all extract groups could significantly reduce the MDA content and increase the activity of SOD,CAT,GSH-Px,the difference was statistically significant(P<0.05).The research results showed that the extract of Radix Rehmanniae Preparata had a good protective effect on the lens of light-damaged mice,and it could relieve visual fatigue.
