中文摘要: 通过离体和活体筛选,确定一株生防菌KD-16J对果实灰霉病的抑制效果。经形态学鉴定、生理生化试验和内源转录间隔区序列分析鉴定,确定其为膜醭毕赤酵母菌(Pichia membranifaciens)属。在平板抑菌试验中发现,在105CFU/mL~108CFU/mL之间,酵母菌的应用浓度越高,对灰葡萄孢菌的抑制效果越好,1×108CFU/mL的生防菌对灰葡萄孢菌菌丝生长的抑制率为85.53%,同时能够完全抑制孢子的萌发;显微镜观察发现与酵母菌共培养能够导致灰葡萄孢菌菌丝肿胀,内部物质泄漏。活体试验(番茄果实)结果表明,该菌株能够显著抑制番茄果实的自然发病率和有伤接种灰葡萄孢菌果实的灰霉病发病率,而且喷洒该菌的时间越早对果实灰霉病的抑制效果越好,当生防菌KD-16J先于灰霉菌36 h处理时,番茄果实的发病率仅为29.12%。
Abstract:The biocontrol bacterium strain KD-16J was found to have an inhibitory effect on fruit gray mold in vitro and in vivo.And it was identified as Pichia membranifaciens according to morphological,biochemical and internally transcribed spacer gene sequencing.The strain was found that the higher the applied concentration,the better the inhibitory effect on Botrytis cinerea between 105CFU/mL and 108CFU/mL.The inhibition rate of 1×108CFU/mL biocontrol strain on the growth of B.cinerea hyphae was 85.53 %,and it could completely inhibit the germination of spores.When B.cinerea and the yeast cells were co-cultured,swollen tested hyphae,inner substance leakage were observed with a microscope.In addition,the natural incidence of tomato fruits and the incidence of gray mold in the fruits of injured inoculation with B.cinerea could be significantly inhibited by the strain,and the earlier the strain was sprayed,the better the inhibitory effect on fruit gray mold disease in vivo experiments.When the biocontrol bacterium strain KD-16J was preceded by B.cinerea for 36 h,the incidence of tomato fruit only was 29.12 %.
