中文摘要: 志贺氏菌是一类具有强传染性和危害严重的革兰阴性食源性致病菌。该文利用自制的CdTe量子点偶联志贺氏菌单克隆抗体,将志贺氏菌单克隆抗体和羊抗鼠二抗分别喷涂硝酸纤维素膜,并分别作为试纸条检测线和质控线,研制一种可视化检测志贺氏菌的量子点免疫荧光试纸条。该试纸条对肉类实际样品的检测限为1×104CFU/mL,单个样品检测时间只需15 min,对常见的5种食源性致病菌均无交叉反应性。该方法构建的基于量子点免疫荧光试纸条满足快速、可视化、高灵敏检测志贺氏菌需求,可用于肉类食品的快速检测。
Abstract:The Shigella is a kind of gram-negative food-borne pathogenic bacteria with strong infectivity and serious harm.In this study,self made CdTe quantum dots were electrostatic coupled with mouse anti-Salmonella monoclonal antibody.Anti-Salmonella monoclonal antibody and sheep anti-mouse antibody were marked on the membrane as test line and control line,separately.A quantum dot immunofluorescence strip was developed for the visual detection of Shigella.the fluorescent immunostrip exhibited a good sensitivity with a limit of detection of 1×104CFU/mL.The detection time of the proposed fluorescent immunostrip for each sample was only 15 min.The fluorescent immunostrip was used to detect five common foodborne pathogens,and the results showed that there were no cross-reactions.The prepared fluorescent immunostrip could provide a visual and highly accurate method for the detection of Shigella in meat.
