中文摘要: 该试验采用超高效液相色谱-四极杆飞行时间质谱联用技术(ultra performance liquid chromatography quadrupole time-of-flight-tandem mass spectrometry,UPLC-Q-TOF-MS/MS),进行南酸枣皮果酒的化学成分指认。运用紫外分光光度法,测定南酸枣皮果酒中总黄酮、总原花青素和总酚的含量。并评价了南酸枣皮果酒体外抗氧化和抑制α-葡萄糖苷酶活性。通过与部分对照品比对,结合液相色谱-串联质谱联用法(liquid chromatography-tandem mass spectrometry,LC-MS/MS)的质谱碎片信息,并参考文献数据从南酸枣皮果酒中指认了34个化合物。结果表明,南酸枣皮果酒中主要含黄酮苷、原花青素低聚体、酚类、有机酸和酯类等成分。总黄酮、总酚和总原花青素的含量分别为15.239、7.595、4.015mg/mL。南酸枣皮果酒清除DPPH自由基和抑制α-葡萄糖苷酶的IC50值分别为2.656、10.996 mg/mL,说明南酸枣皮果酒有优越的体外抗氧化和降血糖活性,同时也表明了南酸枣皮果酒具有优越的保健功能。研究结果旨在为南酸枣皮果酒等高附加值产品的开发和南酸枣果皮废弃物的综合利用提供数据支撑,为类似废弃物的综合利用提供研究思路。
Abstract:Ultra performance liquid chromatography quadrupole time-of-flight-tandem mass spectrometry(UPLC-Q-TOF-MS/MS)was used to identify the chemical components of Choerospondias axillaris fruit peel wine.The contents of total flavonoids,total proanthocyanidins,and total phenols in C.axillaris fruit peel wine were determined by ultraviolet spectrophotometry.Antioxidant and α-glucosidase inhibition activity experiments were also carried out in vitro.By comparing with some reference substances,liquid chromatography-tandem mass spectrometry(LC-MS/MS)fragment information and literature data,a total of 34 compounds were identified in C.axillaris fruit peel wine.The results showed that C.axillaris fruit peel wine mainly contained flavonoid glycosides,proanthocyanidin oligomers,phenols,organic acids and esters.The contents of total flavonoids,total phenols,and total procyanidins were 15.239,7.595,4.015 mg/mL,respectively.The IC50values of C.axillaris fruit peel wine in scavenging DPPH free radical and inhibiting α-glucosidase were 2.656,10.996 mg/mL,respectively.The results showed that C.axillaris fruit peel wine had good antioxidant and α-glucosidase inhibition activities,suggesting C.axillaris fruit peel wine had better health care function.The results of this study provided data support for the development of high value-added products such as C.axillaris fruit peel wine and the comprehensive utilization of C.axillaris fruit peel waste.It also provided idea for the comprehensive development and utilization of similar traditional Chinese medicine waste.
