中文摘要: 采用十二烷基磺酸钠-聚丙烯酰胺凝胶电泳(sodium dodecyl sulfate-polyacrylamide gel electrophoresis,SDSPAGE)、二维电泳和基质辅助激光解析电离飞行时间质谱(matrix assisted laser desorption ionization time-of-flight mass spectrometry,MALDI-TOF/TOF/MS)等技术手段揭示驴骨素蛋白的性质和组成,为具有特征生理活性的驴骨生物活性肽的深层次开发提供依据。结果表明,驴乳粉蛋白含量为47.82 g/100 g,分子量在10 kDa~250 kDa间呈弥散型分布,等电点均匀分布在4.5~6.5之间。通过与马科蛋白图库进行对比,检索出89种蛋白,对这些蛋白进行基因本体论(gene ontology,GO)分类和京都基因与基因组百科全书(kyoto encyclopedia of genes and genomes,KEGG)信号通路富集,发现这些蛋白参与间充质迁移、氨基酸刺激的细胞反应、核小体组装、视网膜稳态、软骨内骨形态发生中的软骨发育等生物过程,也参与血小板激活、黏附斑、蛋白质的消化与吸收等信号通路。
Abstract:Sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE),two-dimensional electrophoresis and matrix assisted laser desorption ionization-time of fight-mass spectrometry(MALDI-TOF/TOF/MS)were used to reveal the properties and composition of donkey bone extract,which provided a basis for the deep development of donkey bone bioactive peptides with characteristic physiological activities.The results showed that the protein content of donkey milk powder was 47.82 g/100 g,the molecular weight was distributed in the range of 10 kDa-250 kDa,and the isoelectric point was uniformly distributed between 4.5-6.5.By comparing with the equine protein library,89 proteins were retrieved,and these proteins were classified by gene ontology(GO)classification and kyoto encyclopedia of genes and genomes(KEGG)signaling pathway enrichment.It was found that these proteins were involved in biological processes such as mesenchyme migration,cellular response to amino acid stimulus,nucleosome assembly,retina homeostasis,cartilage development involved in endochondral bone morphogenesis,and also participate in signal pathways such as platelet activation,focal adhesion,protein digestion and absorption.
