中文摘要: 雨生红球藻细胞培养密度对于虾青素高含量具有重要影响。通过单因素试验考察碳源乙酸钠、pH值、接种量等因素的影响;对发酵工艺的培养时间、碳源乙酸钠含量、初始pH值和接种量4个因素采用L9(34)正交法进行优化试验;对培养基中 4种植物激素 3-吲哚丁酸 (3-indolebutyric acid,3-IBA)、2,4-二氯苯氧乙酸 (2,4-dichlorophenoxyacetic acid,2,4-D)、萘乙酸(1-naphthylacetic acid,NAA)、6-苄氨基嘌呤(6-benzyl aminopurine,6-BA)的添加量进行优化试验。试验结果表明:优化后的发酵工艺为培养时间11 d、接种量30%、初始pH7.5、乙酸钠添加量10 mmol/L,添加植物激素可以提高细胞密度,激素 3-IBA、2,4-D、NAA、6-BA 的最优添加量分别为 0.5、0.1、0.01、0.1 mg/L。优化后雨生红球藻的细胞密度可达1.17×106个/mL,比优化之前提高28倍。
Abstract:Cell density in cultivation of Haematococcus plivialis is important to astaxanthin yield of this alga.The effect and optimization of culture time,sodium acetate,pH and inoculation quantity on cell density of H.pluvialis was investigated by single factor experiments and L9 (34)orthogonal experiments.The optimal addition quantity of four plant hormones,3-indolebutyric acid(3-IBA),2,4-dichlorophenoxyacetic acid(2,4-D),1-naphthylacetic acid(NAA) and 6-benzyl aminopurine(6-BA)was investigated by L9(34) orthogonal experiments.The results demonstrated that highest cell density was got in the followed condition,algae culture time was 11 days,the inoculation quantity was 30%;the initial pH of medium was 7.5 and the addition of sodium acetate in medium was 10 mmol/L.The optimal concentration of 3-IBA,2,4-D,NAA and 6-BA was 0.5,0.1,0.01 mg/L and 0.1 mg/L,respectively.Cell density of H.pluvialis reached 1.17×106cell/mL in the best condition,which was 28 times higher than that before optimization.
