超声辅助提取花生根多酚工艺优化及组成分析

中文摘要: 该研究以花生根为主要试验材料,以乙醇溶液为提取溶剂,采用超声辅助提取花生根中的游离态多酚,并用高效液相色谱法(high performance liquid chromatography,HPLC)分析花生根中多酚的组成及含量分布。检测条件:流动相:A为甲醇,B为0.5%甲酸,采用梯度洗脱程序。研究结果表明:超声辅助提取花生根中多酚的最佳工艺条件为乙醇体积分数70%、料液比1∶30(g/mL)、提取温度45℃、超声时间50 min,获得花生根中多酚的最佳提取量为1.629 mg/g。经高效液相色谱法检测出花生根中主要含有的多酚类物质有茶多酚、白藜芦醇、p-香豆酸、咖啡酸。
Abstract:In this study,free polyphenols in floral roots were extracted by ultrasonic assisted extraction with ethanol solution as the extraction solvent.The composition and content distribution of polyphenols in floral roots were analyzed by high performance liquid chromatography(HPLC).The detection conditions were as follows:mobile phase:A was methanol,B was 0.5% formic acid,and gradient elution procedure was used.The results showed that the optimum conditions for ultrasonic assisted extraction of polyphenols from floral roots were70% ethanol volume,1 ∶30(g/mL)solid-liquid ratio,extraction temperature 45 ℃,and ultrasonic time 50 min,and the average polyphenols content in floral roots was1.629 mg/g.Tea polyphenols,resveratrol,p-coumaric acid and caffeic acid were detected by HPLC.
