中文摘要: 为研究骆驼奶中外泌体对Ⅰ型糖尿病(type 1 diabetes mellitus,T1DM)小鼠血糖和体重的影响,该试验通过超高速离心的方法获得骆驼奶中外泌体,并对C57BL/6J小鼠饲喂高脂高糖饲料以及腹腔注射链脲佐菌素的方法建立Ⅰ型糖尿病(T1DM)小鼠模型。将小鼠随机分为正常对照组、模型组、双胍类药物组和外泌体组,对各组小鼠分别连续40 d灌服生理盐水、盐酸二甲双胍缓释片溶液以及外泌体溶液,并检测血糖浓度以及体重。结果显示,通过超高速离心的方法可以获得完整的外泌体;外泌体可显著降低Ⅰ型糖尿病小鼠的血糖浓度(P<0.01),并能起到恢复小鼠体重的作用。结果表明,骆驼奶中外泌体可以降低血糖,为开发天然纳米药物提供依据。
Abstract:To study the effects of bactrian camel milk exosomes on the blood glucose and body weight in typeⅠdiabetes mellitus(T1DM)mice,exosome was obtained by ultra-high speed centrifugation from camel milk.The model of T1DM mouse was established by fed with high calorie and high sugar diet and intraperitoneal injection streptozotocin (STZ).The mice were randomly divided into normal control group,model group,biguanide group and exosome group.The mice in each group were given normal saline,metformin hydrochloride sustained-release tablet solution and exosome solution for 40 d.Then we detected blood sugar concentration and body weight.The results showed that intact exosomes could be obtained by ultra-high speed centrifugation,exosomes could significantly reduce the blood glucose concentration of typeⅠdiabetic mice mellitus(P<0.01)and could restore the body weight of mice.The results showed that camel milk exosomes could be used as natural nano-grade oral drugs for the treatment of typeⅠdiabetic mellitus.
