中文摘要: 为充分利用蓝靛果,提高其经济价值,该文采用酶解法提高蓝靛果出汁率,研究蓝靛果汁中4种活性物质含量的变化,并用DPPH自由基、·OH、ABTS+自由基的清除率来评价蓝靛果汁的抗氧化能力。应用响应面法对蓝靛果的酶解工艺进行优化,结果表明,最优酶解工艺为果胶酶用量0.27%、纤维素酶用量0.92%、酶解温度47℃。在此工艺条件下,蓝靛果的出汁率为85.67%。酶解对蓝靛果汁中的VC、花色苷、总酚和总黄酮都有积极的影响,酶解后,蓝靛果汁中VC、花色苷和总黄酮的含量分别增加了1.78、1.13 mg/mL和2.54 mg/mL。酶解后的蓝靛果汁对DPPH自由基、·OH和ABTS+自由基的IC50值分别为0.285、6.37、11.62 mg/mL,具有较高的清除能力。当质量浓度为1.8 mg/mL时,蓝靛果汁对DPPH自由基的清除率为88.13%;当质量浓度为12 mg/mL时,蓝靛果汁对·OH的清除率为74.12%;蓝靛果汁在质量浓度为60 mg/mL时,ABTS+·的清除率达到最大为94.67%。
Abstract:In order to make full use of lonicera edulis and increase their economic value,the enzymatic hydrolysis method was studied to improve the juice yield of lonicera edulis and the changes in the content of four active substances in lonicera edulis juice were studied,and the scavenging rates of DPPH·,·OH,ABTS+free radicals were used to evaluate the antioxidant capacity of lonicera edulis juice.The response surface method was used to optimize the enzymatic hydrolysis process of lonicera edulis.The results showed that the optimal enzymatic hydrolysis process was 0.27% pectinase.The cellulose addition amount was 0.92%,and the hydrolysis temperature was 47℃.Under these process conditions,the juice yields of lonicera edulis was 85.67%.After enzymatic hydrolysis,the contents of VC,anthocyanins and total flavonoids in the lonicera edulis juice increased by 1.78,1.13 mg/mL and 2.54 mg/mL.Enzymatic hydrolysis had a positive effect on VC,anthocyanins,total phenols and total flavonoids in lonicera edulis juice.The IC50values of the enzymatically hydrolyzed lonicera edulis juice against DPPH·,·OH and ABTS+free radicals respective were 0.285,6.37 mg/mL and 11.62 mg/mL,and had high scavenging ability.When the mass concentration was 1.8 mg/mL,the clearance rate of DPPH free radicals by lonicera edulis juice was 88.13%.When the mass concentration was 12 mg/mL,the scavenging rate of lonicera edulis juice to·OH radicals was 74.12%.When the mass concentration of lonicera edulis juice was 60 mg/mL,the clearance of ABTS+·reached a maximum of 94.67%.
