响应面法优化超声辅助提取大果青扦叶总黄酮及其抗氧化性研究

中文摘要: 以大果青扦叶为材料,采用超声辅助提取总黄酮,在单因素试验的基础上,采用Box-Behnken设计对大果青扦叶总黄酮的超声辅助提取工艺优化,并研究其抗氧化活性。结果表明最佳提取工艺为:液料比40∶1(mL/g),提取次数2次,超声时间63 min,超声功率480 W,提取温度52℃,乙醇浓度50%,总黄酮提取率为12.12%。在体外大果青扦叶总黄酮对DPPH自由基、ABTS+自由基和亚硝酸盐表现出较好的清除效果,半抑制浓度IC50值分别为1.05、0.35、1.98 mg/mL,表明大果青扦叶总黄酮具有较强的抗氧化能力。
Abstract:The extraction of total favonoids from the leaves of Picea neoveitchii was carried out by ultrasonic assisted extraction (UAE).On the basis of single-factor experiment,the response surface method was used to optimize the ultrasonic extraction process of total flavonoids from Picea neoveitchii leaves,the scavenging rates of DPPH,ABTS+and nitrite radicals were used to study the antioxidant activity of total flavonoids.The results showed that the optimal extraction process was as follows:the liquid-to-material ratio was 40 ∶1(mL/g),the number of extractions was 2 times,ultrasonic time was 63 min,ultrasonic power was 480 W,extractive temperature was 52℃,the ethanol concentration was 50%.Under the above conditions,the extraction yield of total flavonoids was up to 12.12%.The half inhibitory concentration(IC50)for scavenging of DPPH free radical,ABTS+free radical and nitrite were 1.05,0.35,1.98 mg/mL,respectively.The result indicated that the antioxidant activity of total flavonoids from P.neoveitchii leaves was strong in vitro.
