中文摘要: 设计一种基于纳米ZnO材料的快速检测食物中金黄色葡萄球菌的阻抗生物传感器,首先利用多物理场仿真软件(COMSOL)设计出微叉指电极的结构参数,并用电化学方法将纳米ZnO薄膜修饰到电极表面,利用生物素与亲和素间的亲和力将金黄色葡萄球菌抗体固定到电极表面,随后用激光拉曼光谱、循环伏安法、场发射扫描电镜、红外吸收光谱等对传感器进行表征,并用电化学阻抗谱技术对不同浓度菌样进行检测,对检测结果分析得出传感器检测范围为102cfu/mL~107cfu/mL,检出限为43 cfu/mL。试验同时表明传感器特异性强、稳定性高,并且可用于牛奶等食品中金黄色葡萄球菌的检测。
Abstract:A nano-ZnO based impedance biosensor for rapid detection of Staphylococcus aureus in food sample was designed.Firstly,the structural parameters of the micro-interdigited electrode were designed by COMSOL simulation,and the nano-ZnO films were modified on the surface of the electrode by electrochemical method.The affinity between the biotin and avidin was used to immobilize Staphylococcus aureus antibody to the electrode surface,then the sensors were characterized by laser raman spectroscopy,cyclic voltammetry(CV),field emission scanning electron microscope(FESEM)and infrared absorption spectrum,electrochemical impedance spectroscopy(EIS)was used to detect the samples with different bacteria concentrations.The detection range of the sensor was 102cfu/mL-107cfu/mL,and the detection limit(LOD)was 43 cfu/mL.At the same time,the results showed that the sensors had strong specificity and high stability and could be used for Staphylococcus aureus detection in milk and the like.
