中文摘要: 以新疆药桑叶为原料,采用水提醇沉法获得药桑叶粗多糖,利用Sevage法脱除粗多糖中的蛋白质,通过单因素试验、正交试验对药桑叶多糖脱蛋白工艺进行优化,并对脱蛋白前后的多糖进行DPPH自由基抗氧化活性试验。结果表明,药桑叶多糖的最佳除蛋白工艺条件为多糖溶液与Sevage试剂的体积比为1∶2,脱蛋白时间为20 min,脱蛋白次数为3次,蛋白质除去率可达到76.54%,多糖损失率为20.80%,脱蛋白后获得的多糖抗氧化活性比脱蛋白前的活性强。
Abstract:Using Xinjiang Morus nigra leaves as raw material,the crude polysaccharide of Morus nigra leaves was obtained by water extraction and alcohol precipitation method,and the protein in the crude polysaccharide was removed by Sevage method.The deproteinization process of Morus nigra leaves polysaccharide was optimized by single factor test and orthogonal test.The DPPH free radical antioxidant activity test was performed on the Morus nigra leaves polysaccharides before and after deproteinization.The results showed that the optimal protein removal conditions for the polysaccharides of the Morus nigra leaves were that the volume ratio of the polysaccharide solution to the Sevage reagent was 1 ∶2,the deproteinization time was 20 minutes,the deproteinization times were 3 times,and the protein removal rate was 76.54%.The polysaccharide loss rate was 20.80%.The antioxidant activity of the polysaccharide obtained after deproteinization was stronger than that before deproteinization.
