响应面法优化超声波辅助提取山竹果皮多糖的工艺及其体外抗氧活性研究

中文摘要: 以山竹果皮为原料,采用超声波辅助水提法提取其中的多糖。单因素试验证实了各因素影响多糖提取效果的显著程度,在此基础上利用响应面法对提取工艺进行优化,获得最佳工艺参数:液料比49∶1(mL/g)、提取温度46℃、超声时间26 min、超声功率240 W。在此条件下测得多糖提取率为(6.51±0.17)%,与模型预测值6.72%相当。采用体外测试法考察山竹果皮多糖的抗氧化活性。当多糖浓度为1.0 mg/mL时,对羟基自由基(·OH)、超氧阴离子自由基(·O2-)、DPPH自由基的清除率分别可达79.06%、81.57%、79.23%,总还原力达到0.698,且其性能随多糖浓度的增加而增强,说明山竹果皮多糖具有较好的抗氧化活性。
Abstract:Mangosteen peels as starting materials, studies on ultrasound-assisted extraction of polysaccharides were carried out.Based on results of single factored experiments,the extraction process was optimized by response surface methodology,and the best extraction conditions were obtained:liquid-material ratio 49∶1(mL/g),temperature 46℃,ultrasonic time 26 min,and ultrasonic power 240 W,which resulted in the yield of (6.51±0.17)%similar to the expected yield 6.72%of the regression model.Investigations of antioxidant activity revealed that polysaccharides in mangosteen peels had good scavenging activities on radicals.The clearance rates on hydroxyl radical(·OH),superoxide anion radical(·O2-),DPPH radical were 79.06%,81.57%,79.23%respectively when the concentration of polysaccharides reached 1.0 mg/mL,and the total reducing power was 0.698,which were more strong with higher concentration.
