中文摘要: 以丙酮醛修饰β-乳球蛋白产物(methylglyoxal modified β-lactoglobulin product,MG-bLG)为蛋白质结合态晚期糖基化终产物(advanced glycation end products,AGEs)的代表物,通过灌胃摄入方式研究AGEs对小鼠胰岛素抗性的影响。经长时间摄入MG-bLG,相较于对照组(C组),实验组(MG-bLG组)小鼠摄食量增多,体重增大,血清中低密度脂蛋白、总胆固醇和甘油三酯含量显著增高(P<0.05),空腹血糖显著升高(P<0.05)至7.1 mmol/L,空腹胰岛素水平显著升高(P<0.05)至14.8 mU/L,胰岛抵抗指数和胰岛β细胞功能指数同样显著升高(P<0.05)。此外,MG-bLG组小鼠血清中AGEs水平显著升高(P<0.05)至131.99 pg/mL。因此,长期摄入AGEs可改变机体的糖脂代谢情况,促进机体胰岛素抵抗形成,增加机体患2型糖尿病风险。
Abstract:The methylglyoxal-modified β-lactoglobulin product(MG-bLG)was used as a representative of the protein-bound advanced glycation end products (AGEs)to study the effect of protein-bound AGEs on insulin resistance(IR)in mice by intragastric ingestion.After long-term intake of MG-bLG,compared with the control group(group C),the experimental group(MG-bLG group)increased the food intake and weight,the content of serum low-density lipoprotein,total cholesterol and triglycerides were significantly increased (P<0.05),the fasting blood glucose was significantly increased to 7.1 mmol/L (P<0.05),the fasting insulin level was significantly increased to 14.8 mU/L (P<0.05),homeostasis model assessment-insulin resistance index was significantly increased (P<0.05),homeostasis model assessment-insulin sensitivity index was significantly decreased (P<0.05).In addition,the serum AGEs levels in the MG-bLG group were significantly increased to 131.99 pg/mL (P<0.05).Therefore,long-term intake of AGEs can change the glucose-lipid metabolism,promote the formation of insulin resistance,and increase the risk of type 2 diabetes in the body.
