中文摘要: 为有效利用鱼皮制备降血压肽,以罗非鱼皮为原料,采用响应面分析法优化汽爆辅助酶解法制备血管紧张素转换酶(angiotension converting enzyme,ACE)抑制肽工艺条件。结果表明,汽爆辅助酶解制备鱼皮ACE 抑制肽的最优工艺参数为:汽爆压力0.6 MPa、保压时间0.5 min、底物浓度2 %、酶底比0.92 %、酶解时间2 h、pH 值8.4、酶解温度56 ℃。在此条件下鱼皮酶解产物ACE 抑制率理论值为94.97%,实际值为93.16%,其相对分子质量主要分布于307 Da~3 323 Da 之间。本研究结果为鱼皮降血压肽的高效制备、生物活性等方面的研究提供了理论依据。
Abstract:An optimized method using steam explosion-assisted enzymatic hydrolysis was developed for the preparation of angiotension converting enzyme (ACE)inhibitory peptides from fish skin.The response surface analysis method was used to optimize the enzymolysis process parameters based on the single factor experiment.The results showed that the optimum parameters were steam pressure 0.6 MPa,holding for 0.5 min,enzymesubstrate ratio 0.92%,enzymolysis time 2 h,pH 8.4 and temperature 56 ℃.Under the optimal conditions,the theoretical value of ACE inhibition rate was 94.97 %,while the actual value was 93.16 %. Under the optimal conditions,the relative molecular mass of the hydrolysate was concentrated in the range of 307 Da-3 323 Da.The results of this study provide a reference for the preparation of ACE inhibitory peptide from fish skin protein by steam explosion-assisted enzymatic hydrolysis.
