4株羊肚菌胞外多糖含量及其生物活性的研究

中文摘要: 为探讨不同羊肚菌菌株胞外多糖的含量及其抑菌、抗氧化活性和对α-淀粉酶的抑制作用是否有差异,以4 株羊肚菌(编号为YS-Y、XH-0、XH-2、XH-4)为材料,利用醇沉法提取羊肚菌胞外多糖,并用Sevag 法去除蛋白后,测定其胞外多糖含量及其抑菌活性、抗氧化能力和对α-淀粉酶的抑制作用,并将其结果进行对比。结果表明,YS-Y、XH-0、XH-2、XH-4 4 株羊肚菌胞外多糖的含量分别为10.05%、5.80%、19.89%、8.34%;4 株羊肚菌具有一定程度的抑菌能力,菌株XH-2 的抑菌效果优于其它3 株,且对金黄色葡萄球菌和大肠杆菌的抑制效果最好;4 株羊肚菌的还原力、DPPH 自由基、ABTS+自由基以及羟自由基清除率都随着质量浓度的增大而增大,当质量浓度达到1.0 mg/mL时,菌株XH-2 的还原能力和ABTS+自由基清除能力优于其他3 株,其OD700 值和EC50 分别为0.136 和0.56 mg/mL;菌株XH-4 的DPPH 和羟自由基清除率优于其它3 株,其EC50 分别为16.68 mg/mL 和1.40 mg/mL,且4 株羊肚菌的羟自由基清除率均大于VC;菌株XH-2 对α-淀粉酶的抑制作用优于其它3 株,其EC50 为1.55mg/mL。该研究表明羊肚菌胞外多糖具有一定的生物活性,菌株XH-2 和XH-4 具有更大的药用潜力,可为羊肚菌资源开发提供一定的参考意义。
Abstract:The content of extracellular polysaccharides and their antimicrobial,antioxidant and inhibitory effects on alpha-amylase of different Morchella strains was investigated. In this experiment,four strains of Morchella esculenta(YS-Y,XH-0,XH-2,XH-4)were used to extract the extracellular polysaccharides from Morchella esculenta by alcohol precipitation method.After removing the proteins by Sevag method,the content of extracellular polysaccharides,their antimicrobial activity,antioxidant capacity and inhibition of alphaamylase were determined,and the results were compared. The results showed that the contents of extracellular polysaccharides of four morel strains of YS-Y,XH-0,XH-2,and XH-4 were 10.05 %,5.80 %,19.89 %,and 8.34%,respectively;four strains of Morchella had certain bacteriostasis ability.The bacteriostasis effect of strain XH-2 was better than that of other three strains,and had the best bacteriostasis effect on Staphylococcus aureus and Escherichia coli. Four strains of Morchella had the ability of reducing,DPPH free radical,ABTS+free radical and hydroxyl radical scavenging.The rate increased with the increase of mass concentration. When the concentration of XH-2 reached 1.0 mg/mL,the reduction ability and ABTS+free radical scavenging ability of strain XH-2 were better than those of other three strains,with OD700 and EC50 of 0.136 and 0.56 mg/mL respectively;DPPH and hydroxyl radical scavenging rate of strain XH-4 were better than those of the other three strains,with EC50 of 16.68 mg/mL and 1.40 mg/mL respectively,and hydroxyl radical scavenging rate of four strains of Morchella XH-2 was higher than VC;inhibition of strain XH-2 on alpha-amylase was stronger than others,its EC50 was 1.55 mg/mL. The research showed that the extracellular polysaccharides of Morchella esculenta had certain biological activity,and strains XH-2 and XH-4 had greater medicinal potential,which could provide some reference for the development of Morchella esculenta resources.
