中文摘要: 该研究以百香果作为番茄红素提取的原材料,设置单因素试验和正交试验,从而研究其最佳的提取工艺。单因素试验设置5 个因素,分别为:预处理方式、提取剂、提取时间、提取温度、液料比;在单因素试验结果的基础上,从中选取3 个因素(提取时间、提取温度、液料比)设计正交试验。结合单因素试验和正交试验结果,研究表明:影响番茄红素提取量的因素中,液料比为主要影响因素,提取时间和提取温度为次要影响因素且影响效果相近;最优的预处理方法为浆果破碎后15 000 r/min 离心15 min,最佳提取工艺为:丙酮-正己烷2 ∶1(体积比)做提取剂,液料比为3 ∶1(mL/g),在 35 ℃恒温振荡培养箱中提取 3 h。
Abstract:The purpose of this experiment was to provide further data reference and process optimization for the extraction of lycopene.In this experiment,passion fruit(Passiflora edulia Sims)was used as the raw material of lycopene extraction,single factor test and orthogonal test were set up to compare and analyze the two experiments,so as to study the best extraction technology.Five factors were set up in single factor experiment:pretreatment method,extraction agent,extraction time,extraction temperature,liquid-to-material ratio.Based on the results of single factor test,three factors (extraction time,extraction temperature,liquid-to-material ratio)were selected to design orthogonal test.Through the comparison and analysis of single factor test and orthogonal test,the best pretreatment method was centrifugation in 15 000 r/min high speed centrifuge for 15 min,the optimum extraction technology was among the factors affecting the amount of lycopene extraction,the ratio of liquid to material was the main one,the extraction time and temperature.acetone-n-hexane 2 ∶1(volume ratio)as extracting agent,liquid-to-material ratio of 3 ∶1(mL/g).The extract was extracted for 3 h in a constant temperature oscillation incubator at 35 ℃.
