中文摘要: 为探究不同浸泡时间对金银花水提液中有机酸类成分的影响,金银花浸泡0、30、60、90 min,80、95 ℃提取,采用高效液相色谱法测定7 种有机酸类成分的含量变化。结果表明绿原酸在浸泡30 min 后提取效果最好,獐牙菜苷在浸泡90 min 后提取效果最好,隐绿原酸、异绿原酸A、异绿原酸C 均为:直接提取>浸泡30 min>浸泡90 min 和浸泡60 min,绿原酸、新绿原酸、隐绿原酸、异绿原酸B、异绿原酸C 在95 ℃提取效果比80 ℃时好,异绿原酸A 在80 ℃下提取效果比95 ℃时好。结果表明,浸泡对金银花中各有机酸成分均有不同程度的影响,浸泡30 min 后95 ℃提取90 min 的提取条件既能保证金银花中较高的有机酸提取量又能保证生产效率,适当的浸泡能够加快提取进程,提高得率,降低加热提取长时间的耗能。
Abstract:In order to explore the effects of different soaking time on the organic acid components in the Lonicera japonica Flos aqueous extract,the Lonicera japonica Flos was soaked at 0,30,60,90 min and extracted at 80,95 ℃.The content of the seven active ingredients was determined by high performance liquid chromatography(HPLC).The extraction effect of chlorogenic acid was the best after soaking for 30 min.The extraction effect of sweroside was the best after soaking for 90 min.The extraction effects of cryptochlorogenic acid,isochlorogenic acid A and isochlorogenic acid C were as follows:direct extraction > soak for 30 min >soak for 90 min and soak for 60 min.The extraction effect of chlorogenic acid,neochlorogenic acid,cryptochlorogenic acid,isochlorogenic acid B and isochlorogenic acid C at 95 ℃ was better than that at 80 ℃.Isochlorogenic acid A was better at 80 ℃ than at 95 ℃.The results showed that soaking had different effects on various organic acid components in Lonicera japonica Flos.The extraction conditions of 90 min extraction at 95 ℃after soaking for 30 min could ensure the higher organic acid extraction amount in the Lonicera japonica Flos,and ensure the production efficiency.Appropriate soaking could speed up the extraction process,increase the yield,and reduce the energy consumption of heating extraction for a long time.
