中文摘要: 对姜黄挥发油诱导人肺癌A549 细胞的凋亡影响及作用机制进行研究。采用二甲基噻唑蓝比色法(methl thiazolyl tetrazolium,MTT)检测姜黄挥发油对A549 的细胞增殖的抑制作用,采用吖啶橙(acridine orange,AO)染色法、流式细胞仪法、划痕和集落形成试验探究姜黄挥发油诱导细胞凋亡的作用机制。结果表明,姜黄挥发油以时间和剂量依赖性关系抑制癌细胞的增殖。姜黄挥发油浓度在100 mg/L 时就能够使细胞结构发生变形,细胞核发生皱缩,同时与空白对照组相比,加入经不同浓度姜黄挥发油处理后的A549 细胞,其早期和晚期凋亡细胞数量均有所增加,其迁移能力也受到了不同程度的抑制。
Abstract:The effect of Curcuma longa L.volatile oil on apoptosis of human lung cancer A549 cells and its mechanism was studied.Adopted by methyl thiazole blue colorimetry (MTT)to detect the inhibitory effect of Curcuma longa L.volatile oil on cell proliferation of A549.The mechanism of apoptosis induced by Curcuma longa L.volatile oil was investigated by acridine orange(AO)staining,flow cytometry,scratch and colony formation experiments.The results showed that Curcuma longa L.volatile oil inhibitd the proliferation of cancer cells in a time-and dose-dependent manner.When the concentration of Curcuma longa L.volatile oil was 100 mg/L,the cell structure could be obviously deformed and the nucleus could shrink and apoptosis.At the same time,compared with the blank control group,the number of apoptotic cells in the early and late stages of A549 cells treated with different concentrations of Curcuma longa L.volatile oil increased,and their migration ability was also inhibited to varying degrees.
