中文摘要: 为研究阿魏菇多糖(Pleurotus ferulae polysaccharides,PfP)的超声提取工艺及体外抗氧化活性,在单因素试验的基础上,采用中心组合设计(box-behnken design,BBD)和响应面方法(response surface methodology,RSM),研究超声时间、超声温度、液料比及提取次数对PfP 提取率的影响,优化PfP 的超声辅助提取工艺;通过测定对Fe3+还原能力和对DPPH 自由基、羟基自由基(·OH)的清除能力,对PfP 的体外抗氧化活性进行研究。结果显示,超声温度、超声时间以及液料比均对PfP 的提取率有显著影响,其中料液比影响最大,超声时间影响最小;最佳工艺条件为:超声温度66 ℃、超声时间27min、液料比29 ∶1(mL/g)、提取2 次,在此条件下PfP 提取率达到14.69%。体外抗氧化试验结果表明,PfP使Fe3+还原产生的最大吸光值为0.54,对DPPH 和·OH 的最大清除率分别为85.61%和56.1%,其抗氧化活性均在一定浓度范围呈剂量正相关效应。
Abstract:The ultrasonic-assisted extraction process for polysaccharides from Pleurotus ferulae polysaccharides(PfP)and its antioxidant activity in vitro were studied in this paper. Based on the single-factor tests,boxbehnken design and response surface methodology (RSM)were employed to evaluate the effects of ultrasonic temperature,ultrasonic time,ratio of water to sample and extraction times on the extraction rate of PfP. The antioxidant activity of PfP in vitro was studied by measuring its reduction ability and scavenging abilities of DPPH free radical and hydroxyl free radical (·OH).The optimal ultrasonic-assisted extraction process of PfP was based on twice extraction for 27 min at 66 ℃ with a ratio of water to dry material of 29 ∶1 (mL/g),which yielded an extraction rate of 14.69 %. The results of antioxidant experiment in vitro showed that the maximum absorbance value of Fe3+reduction was 0.54,and the maximum scavenging rates of DPPH and·OH were 85.61%and 56.1 % respectively. The antioxidant activities of PfP showed a dose-positive correlation in a certain concentration range.
