中文摘要: 以6 种浙东地区市售咸鳓鱼为对象,采用分离培养的方法结合16S rDNA V4 可变区高通量测序较为全面地探究其中微生物多样性,并且分析分离菌株氨基酸脱羧酶基因及常见抗生素耐药基因携带情况。试验共分离获得151 株可培养细菌,16S rDNA 测序将其鉴定至14 个属和23 个种,其中芽孢杆菌数量最多(98 株,64.90%);葡萄球菌其次(20 株,13.25%);嗜冷杆菌次之(13 株,8.61%)。132 株分离菌株携带组氨酸脱羧酶基因,占比高达 87.42%;鸟氨酸脱羧酶基因和赖氨酸脱羧酶基因的携带比例分别为46.36%和45.03%。耐药基因携带率处于较低水平。高通量测序研究结果表明,门水平上,厚壁菌门(Firmicutes)是市售咸鳓鱼中主要优势微生物。碱杆菌属(Alkalibacillus)、慢性芽孢杆菌属(Lentibacilus)、狭义梭菌属(Clostridium sensu stricto)、梭菌属(Clostridiaceae)等分别在不同样品中占据较高比例,不同来源样品中微生物多样性呈现出较大的差异。
Abstract:Microbial diversity of 6 different slated Chinese herring samples were studied based on isolation and 16S rDNA sequencing,as well as 16S rDNA V4 variable region high-throughput sequencing.Meanwhile,distribution of amino acid decarboxylase gene and antibiotic resistant gene were detected among all the isolates.In total,151 cultivable isolates were picked up,and according to 16S rDNA sequencing results they were classified into 14 genus and 23 species.Among them,Bacillius spp.had the highest number (98 isolates,64.90%),followed by Staphylococcus spp.(20 isolates,13.25%)and Psychrobacter spp.(13 isolates,8.61%).Histidine decarboxylase was detected in 132 isolates (87.42 %),while 46.36 % and 45.03 % of isolates contains ornithine decarboxylase gene and lysine decarboxylase gene respectively.Antibiotic resistant gene carrying rate was at a low overall.16S rDNA V4 variable region high-throughput sequencing results revealed that Firmicutes was the predominant phylums,while Alkalibacillus,Lentibacilus,Clostridium sensu stricto,Clostridiaceae were predominant genus.Microbial diversity varied a lot among different samples.
