中国6个产地文旦柚皮精油化学成分及生物活性研究

中文摘要: 运用气相色谱与质谱联用(gas chromatography-mass spectrometry,GC-MS)分析中国6 个产地文旦柚皮精油化学成分;采用微量肉汤稀释法测试6 种精油对3 种革兰氏阴性菌(大肠杆菌、鼠伤寒沙门氏菌、福氏志贺菌)和3 种革兰氏阳性菌(枯草芽孢杆菌、四联球菌、金黄色葡萄球菌)抗菌活性;选用人体肝癌细胞株和宫颈癌细胞株测试6 种精油细胞毒活性。结果表明,6 个产地文旦柚皮精油共检测出54 种化合物,其中44 种为共有化合物,主要化合物为d-柠檬烯(51.06%~57.28%),β-月桂烯(19.16%~27.95%)和诺卡酮(3.48%~5.27%),基于精油的化学信息进行主成分分析和聚类分析,将6 个产地文旦柚分为3 类;6 种精油均显示一定抗菌活性和细胞毒活性。通过对比研究不同产地文旦柚皮精油化学成分和生物活性的差异,为其在功能性食品方面的科学开发提供依据。

Abstract:The chemical compositions of essential oils in Wendan pomelo peels collected from six areas were analyzed by gas chromatography-mass spectrometry(GC-MS).Micro-broth dilution method was used to evaluate the antibacterial activities of the essential oils on three Gram-negative bacteria (Escherichia coli,Salmonella typhimurium,Shigella flexneri)and three Gram-positive bacteria (Bacillus subtilis,Four Synechococcus,Staphylococcus aureus).Human hepatocellular carcinoma cell line and cervical cancer cell line were selected for cytotoxic activity test.The results showed that 54 compounds in total were identified from the six essential oil samples of Wendan pomelo peels,of which 44 were common compounds.The main compounds were dlimonene(51.06%-57.28%),β-myrcene(19.16%-27.95%)and nocardone(3.48%-5.27%).Based on all chemical compounds information,principal component analysis and cluster analysis were used to classify six areas of Wendan into three categories.All the six essential oils showed antibacterial activity and cytotoxic activity.By comparing the differences in chemical composition and biological activity of essential oils of Wendan peel from different areas,scientific suggestion on the application of oils in functional food were provided.
