金鸡毛草叶总黄酮大孔树脂的富集及抗氧化活性研究

中文摘要: 以金鸡毛草叶总黄酮粗提液为原料,用静态吸附试验对7 种大孔树脂进行筛选,选取用最佳大孔树脂进行后续吸附考察,采用单因素试验优选总黄酮的最佳纯化工艺,以维生素C(VC)为对照品,考察金鸡毛草叶总黄酮的抗氧化活性。结果表明最佳大孔树脂为AB-8 型,经单因素和正交试验,树脂动态吸附最佳纯化工艺为上样浓度4 mg/mL、上样体积1.2 BV、上样流速1.5 BV/h、上样pH6、洗脱水用量3.5 BV、洗脱液乙醇的浓度60%,洗脱用量3 BV,在此条件下纯化后总黄酮的保留率为61.1%,纯化后样液纯度提高至5.24 倍;抗氧化活性方面,在干浸膏浓度20 μg/mL~100 μg/mL 范围内对 1,1-二苯基-2-三硝基苯肼自由基(DPPH·)清除率为 23.10%~57.76%,在 100 μg/mL 浓度下对羟基自由基(·OH)的清除率为 3.76%,在 3.6 μg/mL~18.2 μg/mL 浓度范围内对 2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐自由基(ABTS+·)清除率为33.27%~87.90%,表明金鸡毛草叶总黄酮具有一定的抗氧化活性,可为金鸡毛草的深入开发、产业附加值的提高提供参考依据。

Abstract:Seven kinds of macroporous resins were screened by static adsorption test with extract of total flavonoids from leaves of Rooster maocao.The optimum macroporous resins were selected for subsequent adsorption.The optimum purification process of total flavonoids was optimized by single factor test.Vitamin C(VC)was used as the control to investigate the antioxidant activity of the total flavonoids.The results showed that the optimum macroporous resin was AB-8.By single factor and orthogonal test,the optimum purification process of resin dynamic adsorption was as follows:sample concentration of 4mg/mL,sample volume of 1.2 BV,sample flow rate of 1.5 BV/h,sample pH of 6,washing and dehydration dosage of 3.5 BV,eluent ethanol concentration of 60%,elution dosage of 3 BV.Under these conditions,the purity of purified sample increased to 5.24 times for the retention rate of 61.1%.The scavenging rate of DPPH·was 23.10%-57.76%in the range of 20 μg/mL-100 μg/mL for antioxidant activity,and the ABTS+·clearance rate was 33.27%-87.90% in the range of 3.6 μg/mL-18.2 μg/mL,and the clearance rate of·OH was 3.76 % at 100 μg/mL,indicating that the total flavonoids of Rooster maocao had certain antioxidant activity.This test can provide a reference for further development and industrial added value of Rooster maocao.
