中文摘要: 探究硫酸酯化沙棘叶多糖(sulfated seabuckthorn leaf polysaccharide,SSLP)的解酒作用。采用氯磺酸-吡啶法修饰沙棘叶多糖(seabuckthorn leaf polysaccharide,SLP),氯化钡-明胶浊度法及红外光谱法检测其取代情况。瓦勒-霍赫法检测SSLP及SLP对乙醇脱氢酶(alcohol dehydrogenase,ADH)的影响作用。使用酒精建立小鼠醉酒模型,记录小鼠醉酒潜伏时间、持续醉酒时间,测定血清中ADH活性。结果表明,SSLP在1 244.07、808.16 cm-1处有S=O、C-O-S吸收峰,表明酯化成功,硫酸基取代度为1.63;与模型组比较,SLP和SSLP组小鼠醉酒潜伏期显著延长,持续醉酒时间显著缩短,ADH活性显著增强,且SSLP优于SLP。说明SSLP与SLP均具有较好的解酒作用且SSLP的解酒活性更优,因此可通过衍生化的方式增强SLP的解酒活性。
Abstract:The anti-alcohol mechanism of sulfated seabuckthorn leaf polysaccharide (SSLP)was explored.Sulfated polysaccharide from seabuckthorn leaf polysaccharide(SLP)was derived by using the chlorosulfonic acid-pyridine method.The content of sulfate radical was determined by barium sulfate turbidity.The polysaccharide esterification was confirmed by infrared spectroscopy.The Valle&Hoch method was used to detect the influence of SSLP and SLP on alcohol dehydrogenase(ADH)activity.Mouse drunk model was established by eating alcohol.The latent time of drunkenness and the durative time of drunkenness were recorded.The activities of ADH in serum were determined.The infrared spectrum(IR)spectra showed that the characteristic absorption peaks of S=O and C-O-S appeared near 1 244.07 cm-1and 808.16 cm-1in SSLP.Compared with the model group,positive group,SLP group and SSLP groups could increase sleep latency significantly and reduce the drunken sleep time significantly,increase the content of ADH markedly and SSLP exhibits stronger anti alcohol effect than SLP.It was indicated that both SSLP and SLP have good anti alcohol effect and SSLP hangover activity was better,so the biological activity of SLP could be enhanced by derivatization.
