中文摘要: 探讨鹰嘴豆醇提物对糖尿病小鼠的降血糖效应。将鹰嘴豆醇提物[低、中、高剂量分别为200、400、800 mg/(kg·d·BW)]及阳性对照二甲双胍[227.5 mg/(kg·d·BW)]分别灌胃链脲佐菌素(streptozocin,STZ)诱导的糖尿病小鼠。4 周后测定小鼠血糖、血清胰岛素水平、血清氧化-抗氧化水平,并观察肝脏、胰腺组织病理变化。鹰嘴豆醇提物低、中、高剂量组小鼠血糖水平分别为模型组的83.4%、60.0%、67.9%,以中剂量组降血糖效果最为显著(P<0.05);该组胰岛素水平、胰岛素抵抗指数与胰岛素敏感指数分别是模型组的39.74%、31.09%与1.22 倍(P<0.05),与阳性对照组无明显差异;中剂量组总超氧化物歧化酶(total-superoxide dismutase,T-SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)、过氧化氢酶(catalase,CAT)活性及丙二醛(malondialdehyde,MDA)含量分别是模型对照组的1.09、1.03、1.36 倍及76.4%(P<0.05),且该组小鼠肝脏与胰腺组织形态与结构趋于恢复正常。
Abstract:The anti-diabetes effects of the chickpea ethanol extracts was studied.Streptozocin-induced diabetic mice were intragastricly administrated with different doses of chickpea ethanol extracts [low dose 200 mg/(kg·d·BW),middle dose 400 mg/(kg·d·BW),high dose 800 mg/(kg·d·BW)]and positive group[melbine:227.5 mg/(kg·d·BW)].Four weeks later,diabetic mice's serum glucose level,insulin level,antioxidative enzymes activities were determined and pathological changes of liver and pancreas were observed by hematoxylin and eosin (HE)staining.The serum glucose levels of chickpea ethanol extract groups at low,middle,and high dose were only 83.4%,60.0%,and 67.9%of those in the model group,respectively (P<0.05) and the middle dose group had the most significant hypoglycemic effects.The serum insulin levels,insulin resistance index,and insulin sensitivity index of middle dose group were only 39.74%,31.09%,and 1.22 times of those in the model group(P<0.05);and no significant differences of the above three indexes were found between middle dose group and the positive group (P >0.05).Moreover,in the middle dose group,activities of total-superoxide dismutase(T-SOD),glutathione peroxidase(GSH-Px),catalase(CAT),and malondialdehyde (MDA)content were 1.09 times,1.03 times,1.36 times,and 76.4% of those in the model group(P<0.05);and the morphology and structure of liver cells and islet cells tended to be normal.
