中文摘要: 探讨以罗布麻叶、山楂、决明子为主要成分的保健食品在辅助降脂方面的作用及机制。将罗布麻叶、山楂、决明子提取物进行工艺优化,制成罗布麻叶降脂胶囊,根据保健食品的评定办法,复制大鼠混合型高脂血症模型,观察混合型高脂血症大鼠体重、肝重变化及在30 d 和45 d 其血清中血脂变化情况;同时,利用蛋白免疫印迹(Western blot)手段,检测组织内含半胱氨酸的天冬氨酸蛋白水解酶(Caspase),包括了Caspase-9、Caspase-3、Cleave Caspase-3蛋白表达量。结果表明:经工艺优化后,浓缩后制备稠膏重49 g,加入12.6 g 的糊精,干燥粉碎后重43.8 g,水分为3.04%,制成罗布麻叶降脂胶囊;罗布麻叶降脂胶囊可改善高脂血症引起的大鼠体重、肝重系数、血脂指标及组织形态异常。同时,罗布麻叶降脂胶囊可能通过调节Caspase 家族蛋白表达水平,发挥改善混合型高脂血症作用。
Abstract:The role and mechanism of health foods with Apocynum venetum,hawthorn and cassia as main components in assisting lipid-lowering was explored.Apocynum venetum,hawthom and cassia seed extracts were optimized to prepare Apocynum venetum lipid-lowering capsules.According to the evaluation method of health food,the rat mixed hyperlipidemia model was duplicated to observe the changes in body weight,liver weight and serum lipids of rats with hyperlipidemia at 30 and 45 days.Besides,Cysteine-containing aspartate proteolytic enzymes(Caspase),including Caspase-9,Caspase-3,and Cleave Caspase-3 protein expression,were detected by the method of Western blot.The results showed that after the process optimization,the concentrated paste was concentrated to a weight of 49 g,12.6 g of dextrin was added,and the weight was 43.8 g after drying and pulverization,and the water content was 3.04 %,and the Apocynum venetum lipid-lowering capsule was prepared.Capsules could improve the body weight,liver weight coefficient,blood lipid index and tissue morphology abnormalities induced by hyperlipidemia.At the same time,Apocynum venetum lipidlowering capsule might play a role in improving mixed hyperlipidemia by regulating the expression level of Caspase family proteins.
