海洋源乳杆菌抑菌物质的研究

中文摘要: 为判定从海鱼肠道分离出的乳酸菌产生的主要抑菌物质,从4种海鱼肠道中分离出的41 株菌株中筛选出16株过氧化酶试验阴性、革兰氏染色阳性的杆菌,初步认定为乳杆菌属细菌。以大肠杆菌为指示菌,采用牛津杯琼脂扩散法研究16株杆菌对大肠杆菌的拮抗效果,筛选出抑菌性能较强的6株杆菌,菌株的抑菌圈直径分别为13.73、14.77、13.71、12.84、13.63 mm 和13.65 mm,6株菌编号为S2-4、S2-6、Y3-1、H2-4、B2-4 和B2-10。6株菌对金黄色葡萄球菌、单增李斯特菌、志贺氏菌、副溶血性弧菌和沙门氏菌都有不同程度的抑制作用,其中S2-6 和Y3-1 抑菌效果最强,对所有指示菌的抑菌圈直径都超过14 mm。通过试验最终判定出6株杆菌的抑菌物质为酸或细菌素。经过16S rDNA全长序列分析,最终将菌株B2-4、H2-4、S2-4鉴定为米酒乳杆菌(Lactobacillus sakei),菌株S2-6 和Y2-1 被鉴定为植物乳杆菌(Lactobacillus plantarum)。

Abstract:In order to determine the main bacteriostatic substances produced by the lactic acid bacteria isolated from the intestinal tract of the sea fish,sixteen strains of peroxidase-negative and Gram-positive bacilli were screened from 41 strains isolated from the four marine fish intestines,and were initially identified as Lactobacillus. Using Escherichia coli as indicator bacteria,the antagonistic effect of 16 strains on Escherichia coli was studied by Oxford cup agar diffusion method. Six strains of bacteria with strong antibacterial activity were screened. The inhibition zone diameters of the strains were 13.73,14.77,13.71,12.84,13.63 mm and 13.65 mm,respectively. 6 strains were numbered S2-4,S2-6,Y3-1,H2-4,B2-4 and B2-10. Salmonella,Staphylococcus aureus,Listeria monocytogenes,Vibrio parahemolyticus and Shigella Castellani were used as the indicator bacteria to research the inhibitory effects of referred 6 strains. The result demonstrated that all of the 6 screened strains showed antimicrobial activities in different degree,which the inhibitory zone diameters were more than 14 mm and the inhibitory effects of S2-6 and Y3-1 were the most significant.The results showed that antibacterial substances of the 6 strains would be acid or bacteriocin.The strain B2-4,H2-4 and S2-4 were identified as Lactobacillus sakei by the analysis of 16S rDNA sequence,while the strain S2-6 and Y3-1 were identified as Lactobacillus plantarum.
