中文摘要: 利用低大米蛋白制备过程发酵水解液中富含大米蛋白和肽。以乳杆菌L2-18+风味蛋白酶/菠萝蛋白酶+胃蛋白酶/酸性蛋白酶分步获得的大米蛋白水解液为研究对象,通过超滤获得小于3 kDa 蛋白肽。经阳离子交换、凝胶柱层析和高效液相色谱(high performance liquid chromatography,HPLC)三步分离纯化,确定出高活性ACE抑制肽组分,IC50值为33.81 μg/mL。经液质联用(liquid chromatograph-mass spectrometer,LC-MS)鉴定分析,其多肽序列为VVFFAAAL。
Abstract:Removal solution for the preparation of low protein rice for kidney disease patients is rich in rice protein and peptides. This paper mainly investigated the step-by-step removal of rice by Lactobacillus plantarum 2-18+Flavor protease/bromelain+pepsin/acid proteasen and the removal solution less than 3 kDa was obtained by ultrafiltration.The protein removal solution was separated and purified by cation exchange,gel column chromatography and high performance liquid chromatography(HPLC)three-step separation and the highest ACE inhibitory activity components in the removal solution was identified by liquid chromatograph-mass spectrometer(LC-MS).The peptide sequence was VVFFAAAL,which IC50 value was 33.81 μg/mL.
