中文摘要: 为提高赤红球菌产红色素的能力,以实验室保藏的赤红球菌(Rhodococcus ruber YM-2)为出发菌株,采用等离子体单独诱变、等离子体连续诱变、紫外线单独诱变、紫外线等离子体复合诱变等方法,最终在等离子体连续诱变处理分别45、30 s时,经筛选得到一株遗传稳定的突变株A2,红色素粗品产量达到1.12 g/L,相比于出发菌株提高了30.32%。同时发现等离子连续诱变比等离子体、紫外线单独诱变,或紫外线和等离子复合诱变的正突变率高,达到39.40%,等离子体连续诱变可作为一种高效的诱变手段。
Abstract:In order to improve the ability of Rhodococcus ruber to produce red pigment,an original strain Rhodococcus ruber YM-2 preserved by our laboratory was used to be screened with plasma mutation,plasmaplasma continuous mutation,ultraviolet mutation,ultraviolet-plasma compound mutation.Under the condition of plasma-plasma continuous treatment at 45,30 s,a genetically stable mutant A2was screened by screening plate and flask fermentation culture.The red pigment yield of A2was 1.12 g/L,increased by 30.32%compared with that of the original strain YM-2.In addition,comparing the effect of plasma,ultraviolet and ultravioletplasma treatments on positive mutation rate,the positive mutation rate of plasma-plasma continuous mutation wasthehighest39.40%,plasma-plasma continuous mutation could be used as a highly effective mutation method.
