中文摘要: 采用水提醇沉法从黑木耳粉中提取黑木耳多糖。通过离子交换纤维素(Diethylaminoethyl-52,DEAE-52及葡聚糖凝胶层析Sephadex G-100对黑木耳子实体多糖(Auricularia auricula polysaccharide,AAP进行纯化,采用红外光谱(infrared spectroscopy,FT-IR、高效液相色谱(high performance liquid chromatography,HPLC、气质联用(gas chromatography-mass spectrometry,GC-MS技术对其主要组分的理化性质和化学结构进行分析,结果表明:AAP为一种具有高分支结构的葡聚糖,黏均分子量为4.50×103Da,其所含鼠李糖、葡萄糖、木糖、甘露糖和半乳糖的含量比分别为0.3%、17.1%、0.3%、3.1%和63.2%,通过进一步对AAP甲基化分析,并对其糖残基的连接方式进行分析,AAP含有→6-α-D-Galp-(1→、-Manp-(1→、→3,4-α-D-Manp-(1→、→3-α-D-Manp-(1→和→6-α-D-Glcp-(1→等残基。
Abstract:Auricularia auricula polysaccharide (AAP)was extracted from Auricularia auricula powder with extraction and alcohol precipitation,and then purified using ion exchange cellulose(DEAE-52)and Sephadex gel chromatography(Sephadex G-100),the physicochemical properties and chemical structures of main components were characterized using the gas chromatography-mass spectrometry (GC-MS),infrared spectroscopy(FT-IR),high performance liquid chromatography(HPLC).The results showed that AAP was a high branched glucan,the viscosity average molecular weight was 4.50 × 103Da,the percent of rhamnose,glucose,xylose,mannose and galactose was 0.3%,17.1%,0.3%,3.1%,and 63.2%,respectively,the methylation analysis were used to analyse the sugar residue of AAP,the result showed that AAP contained →6)-α-D-Galp-(1→,-Manp-(1→,→3,4)-α-D-Manp-(1→,→3)-α-D-Manp-(1→ and →6)-α-DGlcp-(1→.
