中文摘要: 探讨葛根(Pueraria lobata(Willd.)Ohwi)的乙醇提取物对糖尿病肾病肾功能的保护作用,通过腹腔注射氯化镉联合高脂高糖饲料喂养的诱导方式构建糖尿病肾病小鼠模型,保护组灌胃葛根提取物,模型组和空白组灌胃同体积生理盐水,建模完成后取血清样本和肾脏组织分析。结果发现,与空白组正常小鼠相比,模型组小鼠空腹血糖值明显升高(P<0.001);血清中肌酐(creatinine,CRE)、血尿素氮(blood urea nitrogen,BUN)含量明显增加(P<0.001);乙酰氨基葡萄糖苷酶(N-acetyl-β-glucosaminidase,NAG)活性明显增强(P<0.001),且肾脏组织的苏木精-署红(hematoxylineosin staining,HE)染色病理切片在光学显微镜下可见清晰的病变结构。经灌胃葛根提取物后,与模型组相比保护组小鼠各生理生化指标都有所恢复,切片观察组织病变也相对减轻。说明葛根能够有效缓解糖尿病肾病小鼠的肾损伤过程,维持其肾功能至正常水平,有望开发成为针对糖尿病肾病患者的特殊膳食食品。
Abstract:The protective effect of ethanol root extract of Pueraria lobata(Willd.)Ohwi(PU)on renal function in diabetic nephropathy was investigated.A mouse model of diabetic nephropathy was constructed by intraperitoneal injection of cadmium chloride in combination with high-fat and high-sugar diet.Then the protective group was given the extract of PU,and the model group and the control group were given the same volume of normal saline.After modeling,serum samples and kidney tissue analysis were taken.The results showed that the fasting blood glucose level of the model group was significantly higher than that of the blank group(P<0.001),serum creatinine(CRE)and blood urea nitrogen(BUN)levels increased significantly(P<0.001),and N-acetyl-β-glucosaminidase (NAG)activity was significantly enhanced (P<0.001),and hematoxylin-eosin staining(HE)staining pathological sections of kidney tissue showed clear lesion structure under light microscope.After the rats were given the ethanol root extract of PU,the physiological and biochemical indexes of the mice in the protected group were restored compared with the model group,and the tissue lesions were also relatively relieved.It was proved that PU can effectively alleviate the renal injury process in mice with diabetic nephropathy and maintain its renal function to normal level.Therefore,the ethanol root extract of PU is expected to be developed into a special dietary food for patients with diabetic nephropathy.
