中文摘要: 探究紫球藻多糖对体外免疫细胞免疫功能的影响。用不同浓度的紫球藻多糖刺激小鼠腹腔巨噬细胞RAW264.7,用脂多糖(lipopolysaccharide,LPS)作为阳性对照,格里斯试剂(Griess)法检测细胞上清液中一氧化氮(nitric oxide,NO)释放量,噻唑蓝(3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide,thiazolyl blue tetrazolium bromide,MTT)法测定紫球藻多糖对RAW264.7细胞活力的影响,中性红法检测细胞的吞噬能力。采用酶联免疫吸附法(enzyme-linked immunosorbent,ELISA)检测细胞上清液中肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)与白细胞介素-6(interleukin-6,IL-6)的分泌情况。结果表明:不同浓度紫球藻多糖能够促进NO的释放,增强巨噬细胞对中性红的吞噬能力,刺激TNF-α与IL-6的释放。紫球藻多糖浓度在12.5 μg/mL~400 μg/mL的范围内,对细胞无毒性。试验结果初步证明:紫球藻多糖对巨噬细胞RAW264.7具有免疫调节作用,这将为进一步开发应用紫球藻多糖奠定基础。

Abstract:Abstract:To explore the effect of porphyridium creuntum polysaccharide on immunocell functions in vitro.Different concentrations of porphyridium creuntum polysaccharide was used to stimulate the mouse peritoneal macrophage RAW264.7.Lipopolysaccharide(LPS)was used as a positive control.The release of nitric oxide(NO)was detected by griess regent.Cell vitality was determined by MTT method and phagocytic ability was tested by neutral red.The secretion level of tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6)was detected by enzyme-linked immunosorbent(ELISA).Results showed that different concentrations of polysaccharide could promote the production of NO,and enhance the phagocytosis ability of microphage RAW264.7.The polysaccharide could also stimulate the secretion of TNF-α and IL-6.It was no cytotoxicity when the concentration of porphyridium creuntum polysaccharide was at the rang of 12.5 μg/mL-400 μg/mL.This research preliminary prove that the polysaccharide had immunomodulatory effect on macrophages RAW264.7.This effect on macrophages RAW264.7 would be the basis for further development and application of creuntum polysaccharide polysaccharide.
