大蒜多糖对急性酒精性肝损伤小鼠肠道菌群失调的影响

中文摘要: 应用肠杆菌基因间重复共有序列基因扩增(enterobacterial repetitive intergenic consensus-polymerase chain reaction,ERIC-PCR)和聚合酶链式反应--变性梯度凝胶电泳(polymerase chain reaction-denaturing gradient gel electrophoresis,PCR-DGGE)研究大蒜多糖对急性酒精性肝损伤小鼠肠道菌群失调的影响。灌胃红星二锅头建立小鼠急性酒精性肝损伤模型,ERIC-PCR和PCR-DGGE电泳获得肠道菌群指纹图谱,分析肠道菌群的相似性和多样性并鉴定优势条带序列。ERIC-PCR结果表明急性酒精性肝损伤模型小鼠肠道菌群结构发生明显改变,以360 bp的条带为优势条带,610 bp左右的条带强度减弱,大蒜多糖预防给药组中360 bp左右的条带强度减弱。PCR-DGGE结果显示急性酒精性肝损伤的小鼠肠道菌群多样性指数降低,优杆菌属和乳酸菌属细菌明显减少。大蒜多糖高剂量组的多样性指数和均匀度指数最高,优杆菌属和乳酸菌属细菌增多。大蒜多糖可以促进肠道中优杆菌属和乳酸菌属生长,调节急性酒精性肝损伤伴有的肠道菌群失调。
Abstract:Abstract:To study the effects of garlic polysaccharide on the intestinal micrflora dysbiosis in acute alcoholinduced hepatic injury(AAHI)mice by using enterobacterial repetitive intergenic consensus-polymerase chain reaction(ERIC-PCR)and polymerase chain reaction-denaturing gradient gel electrophoresis(PCR-DGGE)techniques.AAHI model in mice was built by Erguotou.The diversity,similarity analyses and the sequences of the dominant bands were carried out by ERIC-PCR and PCR-DGGE techniques.ERIC-PCR analysis showed that the intestinal microflora structure was changed in AAHI group,the band of 360 bp was dominant band,and the intensity of band of 610 bp was decreased.In garlic polysaccharide-pretreated group,the intensity of band of 360 bp was decreased.PCR-DGGE analysis showed that Shannon-Weaver indexes decreased in AAHI group,and Eubacterium and Lactobacillus were both decreased obviously.In garlic polysaccharide-pretreated group,Shannon-Weaver indexes and evenness indexes were the highest,and Eubacterium and Lactobacillus were both increased.Garlic polysaccharide can improve the growths of Eubacterium and Lactobacillus,and regulate intestinal microflora dysbiosis associated with alcohol-induced hepatic injury.
