中文摘要: 使用2,6-吡啶二羧酸(DPA)、L-丙氨酸、肌苷等诱导芽孢萌发,从萌发液中提取芽孢皮层裂解酶。提取的芽孢皮层裂解酶经超滤浓缩后进行SDS-PAGE垂直板电泳分析。研究发现:DPA、L-丙氨酸、肌苷以及L-丙氨酸结合肌苷对枯草芽孢杆菌芽孢均有诱导萌发的作用,且DPA诱导枯草芽孢杆菌芽孢萌发效果最好。进行SDS-PAGE电泳时,进样量为15 μL,分离胶浓度为10%的凝胶板跑出的蛋白条带分离的较好,凝胶板上有4条蛋白条带,通过蛋白质分子量标准曲线可得出,目标蛋白分子量大约分别为:61.10、48.64、43.25、31.28 kDa。本研究初步提取纯化了皮层裂解酶,为后继进一步提纯皮层裂解酶、研究HPTS对该酶活性和结构的影响提供试验材料,有助于阐明高压热处理下芽孢死亡的机理,推动HPTS技术在食品工业中的应用。
Abstract:Spore germination was induced by 2,6-pyridinedicarboxylic acid (DPA),L-alanine and inosine,and the cortex-lytic enzyme was extracted from the germinated spores suspension.The cortex-lytic enzyme was analyzed by SDS-PAGE vertical plate electrophoresis after ultra-filtration.The results indicated that DPA,L-alanine,inosine and L-alanine combined with inosine all induced spore germination efficiently,while the spore germination induced with DPA was the best.During SDS-PAGE gel electrophoresis,the injection volume of 15 μL and 10%separation gel were better for proteins separation,and there were four protein bands on the gel plate.According to the standard curve of protein's molecular weight,the molecular weight of the target proteins were approximately 61.10,48.64,43.25 and 31.28 kDa,respectively.In this study,the cortex-lytic enzyme was extracted and preliminarily purified,which provided experimental materials for the subsequent study on the effect of high-pressure thermal sterilization(HPTS)on the activity and structure of the enzyme.It was helpful to elucidate the mechanism of spore inactivation under HPTS and promote the application of HPTS in food industry.
