中文摘要: 采用单因素试验、正交设计试验、方差分析和多重比较等方法,研究不同因素对田菁叶片蛋白质提取的影响,确定提取的最佳工艺参数。结果表明:影响田菁叶片蛋白质提取率的主次因素依次为提取温度、提取时间、料液比、盐浓度和pH值;提取的最佳工艺参数为:料液比1∶20(g/mL)、pH8、NaCl2%、提取温度60℃,提取时间120 min。在此条件下,田菁叶片蛋白质的提取率可达40.02 mg/g。

Abstract:In order to optimize the conditions for extracting leaf proteins in Sesbania cannabina,single factor tests,orthogonal experimental design,variance analysis and multiple comparisons were applied to study the effects of different factors and optimize the parameters.Results showed that:the five affecting factors for protein yield rank from strong to weak as extraction temperature,extraction time,material/solvent ratio,NaCl concentration,and pH.The optimal extraction conditions were material/solvent ratio 1 ∶20 (g/mL),pH 8,NaCl concentration 2%,extracting time 120 min and extraction temperature 60℃.The yield of protein under the optimal conditions was 40.02 mg/g.
