中文摘要: 以狗枣猕猴桃叶粗多糖得率为响应值,在单因素试验的基础上,采用三因素三水平响应面分析法(RSM),研究超声辅助提取各因素对狗枣猕猴桃叶粗多糖得率的影响,优化超声辅助提取狗枣猕猴桃叶粗多糖最优工艺;利用DPPH自由基清除试验,测定狗枣猕猴桃叶多糖的抗氧化活性。结果表明:超声辅助提取狗枣猕猴桃叶粗多糖最优工艺为超声功率989.61 W,提取时间20.00 min,液固比为80∶1(mL/g),狗枣猕猴桃叶粗多糖得率理论值为5.59%,设定超声功率990 W,提取时间20.00 min,液固比80∶1(mL/g)进行验证试验,狗枣猕猴桃叶粗多糖得率为5.41%。在此条件下得到的狗枣猕猴桃叶多糖具有清除DPPH自由基的能力,清除能力与浓度大小相关。
Abstract:The ultrasonic-assisted extraction technology and antioxidant activities of polysaccharide from the leaves ofActinidia kolomikta(Rupr.et Maxim.)Planch were researched in this paper.Taking the yield of crude polysaccharide as the response value,on the basis of single factor experiment,the effect of ultrasonic-assisted extraction factors on polysaccharide were researched.A three-level-three-factor response surface methodology design was employed to optimize the three extraction variables and to determine the optimum technology.And the antioxidant capacity of the polysaccharide was assessed through DPPH radical scavenging test.The experimental results showed that the optimum technology was that the ultrasonic power was 989.61 W,the extraction time was 20 min,the extraction liquid-to-solid ratio was 80 ∶1(mL/g),under the condition the yield of the crude polysaccharide from the leaves ofActinidia kolomiktawas 5.59%.Setting ultrasonic power as 990 W,extraction time as 20 min and liquid-to-solid ratio as 80 ∶1(mL/g)to verify the optimum extraction technology,and the yield of the crude polysaccharide was 5.41%.The polysaccharide extracted from the leaves ofActinidia kolomiktahad effect of scavenging DPPH radical,and the ability of scavenging DPPH radical was proportional to the concentration of polysaccharide.
