中文摘要: 采用大孔吸附树脂分离石榴皮中安石榴苷。对比吸附率和解吸率,筛选大孔吸附树脂;考察梯度洗脱和等度洗脱分离纯化效果,选择分离方式;通过单因素试验,确定分离纯化参数。试验结果显示:选用LX-3010树脂在长径比为5∶1树脂柱中以等度洗脱方式分离纯化可以得到较高的安石榴苷提取量。单因素结果显示,树脂分离条件为:吸附液浓度2.0 mg/mL,吸附时间30 min,吸附流速1.5 mL/min;洗脱乙醇浓度40%,洗脱时间180 min,洗脱流速1.5 mL/min。在此条件下,可以得到纯度为80.12%的安石榴苷纯化产物。

Abstract:Macroporous adsorption resin was used to separate punicalagin from pomegranate husk.By comparing the adsorption rate and desorption rate,the macroporous adsorption resin was screened;by investigating gradient elution and isocratic elution on effect of separation and purification,separation mode was chosen;by using single-factor test,the parameters for separation confirmed.The result showed that the more punicalagin could be gotten by using LX-3010 resin in column of which the ratio of length to diameter was 5∶1 by isocratic elution.Single-factor test result showed the optimization condition for separation was:the content,the time,and the flow rate for adsorption were 2.0 mg/mL,30 min,and 1.5 mL/min;the content of ethnol,the time,and the flow rate for elution were 40%,180 min,and 1.5 mL/min,respectively.The punicalagin with purity of 80.12%was achieved under the condition.This processing could be a reference for comprehensive utilization of pomegranate husk.
