中文摘要: 采用磷酸缓冲液-乙酸乙酯混合提取、正己烷除脂、MCX 柱净化,采用液相色谱-质谱法(LC-MS/MS)建立了 同时测定猪肉中泰妙菌素及 8-α-羟基泰妙菌素残留量的分析方法。结果表明:在泰妙菌素和 8-α-羟基泰妙菌素 加标浓度 5.0 μg/kg~500.0 μg/kg 范围内,相关系数分别为 0.999 8 和 0.999 7,线性关系良好;检测限(LOD)分别为 1.5 μg/kg 和 3.0 μg/kg,定量限(LOQ)分别为 5.0 μg/kg 和 10.0 μg/kg;平均回收率为 83.4 %~97.6 %,相对标准偏差(RSD)为 2.4 %~5.3 %。
Abstract:Extracting by using phosphate buffer and ethyl acetate mixed, and degreasing by n-hexane, and pu- rifying by MCX column, the method was established for determination of the content of pleuromulin and 8-αhydroxy-pleuromulin in pork by liquid chromatography-mass spectrometry (LC-MS/MS). The results showed that the correlation coefficients of pleuromulin and 8-α-hydroxy-pleuromulin were 0.999 8 and 0.999 7 in spiked concentration from 5.0 μg/kg to 500.0 μg/kg, and they had good linear relationsh. The limit of detection(LOD) were 1.5 μg/kg and 3.0 μg/kg, and the limit of quantification (LOQ)were 5.0 μg/kg and 10.0 μg/kg; the average recovery rate were from 83.4 % to 97.6 %, and relative standard deviations(RSD)were from 2.4 % to 5.3 %.
